| Product Name | RT-LAMP pH-Sensitive Dye Chromogenic Kit (Lyophilizable) |
| Catalog No. | RESK-0110 |
| Description | This RT-LAMP kit integrates a pH-sensitive visual dye directly into the reaction buffer, which contains dATP, dCTP, dGTP, dUTP and other components required for amplification by Bst II DNA Polymerase. The kit includes glycerol-free Bst II DNA Polymerase, uracil DNA glycosylase (UDGase) and a reverse transcriptase mix, and is formulated with a lyoprotectant so the reaction system can be lyophilized directly without further optimization. The reagent delivers high amplification efficiency and sensitivity, and results can be read by the naked eye: positive reactions turn orange-yellow while negative reactions remain magenta, providing a strong color contrast. The kit can also be used directly as a liquid reagent without lyophilization. |
| Intended Use | Rapid visual detection of pathogen RNA by one-step RT-LAMP; suitable for high-throughput screening of large populations. For research use. |
| Principle / Technology | pH-based colorimetric RT-LAMP. A visible pH-sensitive dye serves as the indicator: during isothermal amplification at 60-65 °C, proton release accompanying DNA synthesis lowers the pH of the reaction, driving a distinct color change. Reverse transcription and LAMP amplification are completed in a single step using a thermostated water bath or a standard PCR instrument, and a result can be obtained within 30 minutes. |
| Detection Method | Colorimetric readout by pH-sensitive dye; results are judged visually by color change - positive reactions turn orange-yellow and negative reactions remain magenta. |
| Sample Type | RNA nucleic acid samples (e.g., pathogen RNA extracts) |
| Performance Range / Specifications | High amplification efficiency and sensitivity One-step RNA amplification completed within 30 min at 60-65 °C Clear visual readout with strong contrast between positive (orange-yellow) and negative (magenta) results |
| Reaction Conditions / Protocol | Incubate at 60-65 °C for 30 min (isothermal, one-step RT-LAMP) Heat inactivation at 85 °C for 5 min; read results after the inactivation step is completed |
| Components / Formulation | 2.5x pH-Sensitive Reaction Buffer containing dATP, dCTP, dGTP, dUTP, pH-sensitive indicator and lyoprotectant Glycerol-free Bst II DNA Polymerase UDGase Glycerol-free reverse transcriptase (RT) enzyme mix |
| Storage Conditions | Part I (2.5x pH-Sensitive Reaction Buffer / lyoprotectant): store at -25 to -15 °C Part II (glycerol-free RT enzyme mix / Bst enzyme): store at 2-8 °C |
| Shelf Life | Part I: 6 months at -25 to -15 °C; Part II: 3 months at 2-8 °C |
| Package Specifications | 100 T; 1000 T |
| Product Form | Liquid kit (lyophilizable formulation with lyoprotectant) |
| Shipping Conditions | Part I: shipped on dry ice; Part II: shipped with ice packs |
| Compatibility | Compatible with thermostated water baths and standard PCR instruments; suitable for direct lyophilization |
| Application Notes / Precautions | The kit can be used directly as a liquid reagent without lyophilization Read results only after the 85 °C, 5 min inactivation step, not immediately after the 60-65 °C amplification UDGase in the formulation helps control carry-over contamination from dUTP-containing amplicons |
For research use only, not for clinical use.
