| Product Name | Advanced One-Step RT-qPCR Probe Mix (UDG Plus, 5x) |
| Catalog No. | RESK-0127 |
| Description | This kit is a high-sensitivity, high-concentration (5x) one-tube RT-qPCR premix designed for multiplex quantitative PCR reactions using RNA templates. Reverse transcription and quantitative PCR are carried out in the same reaction tube, simplifying operation and lowering contamination risk. The kit uses a thermostable reverse transcriptase for efficient first-strand cDNA synthesis together with a hot-start Taq DNA polymerase for quantitative amplification. It comprises an optimized MP Buffer and an Enzymes Mix; the buffer already contains Mg2+ and dNTPs along with factors that suppress non-specific PCR amplification and enhance multiplex qPCR amplification efficiency, enabling robust multiplex amplification without compromising primer amplification efficiency. A dUTP/UDG system is included to effectively guard against aerosol contamination. |
| Intended Use | Multiplex probe-based quantitative PCR detection with RNA templates in a convenient one-tube format. |
| Principle / Technology | One-step probe-based RT-qPCR: a thermostable reverse transcriptase efficiently synthesizes first-strand cDNA, followed by quantitative amplification with a hot-start Taq DNA polymerase. The optimized buffer contains Mg2+, dNTPs, factors that suppress non-specific amplification, and factors that boost multiplex amplification efficiency. The dUTP/UDG carry-over prevention system minimizes aerosol contamination risk. |
| Detection Method | Probe-based real-time fluorescent quantitative PCR |
| Sample Type | RNA templates |
| Performance Range / Specifications | 1. Buffer and enzyme components required for RT-qPCR are combined in a single tube for ease of use; 2. Excellent stability: performance is unaffected after 7 days of accelerated storage at 37 °C or 20 repeated freeze-thaw cycles; 3. Includes a dUTP/UDG anti-contamination system. |
| Reaction Conditions / Protocol | One-tube protocol: reverse transcription and qPCR performed sequentially in the same reaction tube; add RNA template, primers, and probes to the 5x mix and run on a real-time PCR instrument. |
| Components / Formulation | Optimized MP Buffer (containing Mg2+ and dNTPs, with factors suppressing non-specific amplification and enhancing multiplex efficiency), Enzymes Mix (thermostable reverse transcriptase and hot-start Taq DNA polymerase), and the dUTP/UDG anti-contamination system. |
