Product Name Universal TaqMan Multiplex qPCR Mix (UDG Plus, with ROX)
Catalog No. RESK-0136
Description This multiplex real-time quantitative qPCR premix is built on a new-generation antibody-based hot-start Taq DNA polymerase. The premix contains the universal components required for qPCR amplification, including dNTPs, Mg2+, Taq DNA polymerase, and UDG; it does not contain gene-specific primers or probes, which should be designed and synthesized by the user. The ROX reference dye is premixed, so no separate addition is required, and the mix is compatible with quantitative PCR instruments requiring either Low ROX or High ROX concentrations. For instruments that do not require ROX normalization, a ROX-free qPCR mix is recommended. This product is suitable for applications requiring ROX dye normalization. Combined with gene-specific primers, probes, and template, it supports a wide range of qualitative and quantitative assays, including detection of residual DNA in intermediates, semi-finished, and finished products of biologics in the biopharmaceutical field, and can be paired with magnetic bead-based residual DNA sample pretreatment kits. It can also serve as a core raw material for research and in vitro diagnostics, for the development of differential detection kits for human bacterial, fungal, and viral pathogens and animal diseases.
Intended Use For qualitative and quantitative TaqMan probe-based qPCR analysis in applications requiring ROX normalization, including residual host-cell DNA quantification in biopharmaceutical products, gene copy number analysis, viral screening, cell quality control, and development of pathogen detection assays.
Principle / Technology Probe-based (TaqMan) multiplex quantitative PCR using a directed-evolution, antibody-modified hot-start Taq DNA polymerase and an optimized buffer system. The built-in ROX reference dye corrects well-to-well fluorescence variation on instruments requiring Low or High ROX normalization. A UDG-based carryover contamination control system is included.
Detection Method TaqMan probe-based multiplex real-time quantitative PCR fluorescence detection with ROX reference dye normalization.
Sample Type DNA templates (e.g., residual host-cell DNA such as E. coli or CHO DNA, pathogen DNA)
Performance Range / Specifications Linear detection range demonstrated with E. coli DNA control from 30 fg/uL to 300 pg/uL and CHO DNA control from 3 fg/uL to 30 pg/uL.
Amplification efficiency 90-110%, R2 > 0.99.
Supports ultra-fast amplification programs of under 30 minutes, significantly reducing overall assay time.
Validated for detection range, limit of quantitation, and precision testing in pharmaceutical customer project types.
Sensitivity / LOD Limit of quantitation (LOQ): 30 fg/uL for E. coli residual DNA and 0.3 fg/uL for CHO residual DNA (confirmed over 10 replicate tests).
Specificity The directed-evolution hot-start polymerase and buffer system suppress non-specific amplification while delivering high sensitivity and good reproducibility.
Reaction Conditions / Protocol Supports ultra-fast amplification programs completed within 30 minutes. Combine the mix with gene-specific primers, probes, and template according to the target assay.
Components / Formulation Premix containing antibody-based hot-start Taq DNA polymerase, dNTPs, Mg2+, UDG, and premixed ROX reference dye.
