Product Name Universal Advanced Multiplex One-Step RT-qPCR Mix (UDG Plus)
Catalog No. RESK-0130
Description The Universal Advanced Multiplex One-Step RT-qPCR Mix (UDG Plus) is a third-generation universal, high-sensitivity, inhibitor-tolerant reagent for multiplex quantitative PCR with RNA templates. Reverse transcription and quantitative PCR are carried out in the same reaction tube, simplifying the workflow and reducing contamination risk. The mix shows excellent universality, delivering outstanding amplification performance with a wide range of primer/probe sets. Its buffer contains factors that effectively suppress nonspecific PCR amplification and factors that boost multiplex qPCR amplification efficiency, improving both the sensitivity and specificity of multiplex assays. A dUTP/UDG carryover prevention system effectively guards against aerosol contamination. The mix also tolerates swab-sample matrices well and supports direct RT-qPCR detection of oral swab samples.
Intended Use For research use in molecular diagnostic assay development: multiplex quantitative detection of RNA targets, including direct RT-qPCR from oral swab samples and multiplex respiratory pathogen panels.
Principle / Technology One-step RT-qPCR: reverse transcription and quantitative PCR proceed in a single closed tube, simplifying operation and reducing contamination risk.
The optimized buffer contains factors that suppress nonspecific amplification and enhance multiplex amplification efficiency, raising multiplex sensitivity and specificity.
Carryover contamination control: a dUTP/UDG system degrades dU-containing carryover PCR products, effectively preventing aerosol-borne amplicon contamination.
Detection Method Probe-based real-time quantitative PCR (one-step RT-qPCR); supports multiplex amplification (validated in quadruplex respiratory panels).
Sample Type RNA templates (validated with serially diluted human RNA); oral swab samples for direct amplification without prior nucleic acid purification.
Performance Range / Specifications Excellent universality: outstanding amplification performance with a wide variety of primer/probe sets.
Fast protocols supported: total detection time of 35 minutes or less.
Enhanced multiplex performance: buffer additives suppress nonspecific amplification and improve multiplex amplification efficiency, increasing sensitivity and specificity.
Built-in dUTP/UDG anti-contamination system effectively prevents aerosol contamination.
Strong inhibitor tolerance: supports direct RT-qPCR detection of oral swab samples; in quadruplex respiratory multi-target assays, direct swab amplification shows no performance loss.
Thermal stability: performance unaffected after 7-day accelerated storage at 37 °C in three product formats.
Sensitivity / LOD High sensitivity demonstrated near the limit of detection: with serially diluted human RNA templates at LOD-level concentrations, the mix delivers superior detection rates and peak shapes compared with comparable products.
Specificity Buffer factors effectively suppress nonspecific PCR amplification, improving multiplex amplification specificity.
Reaction Conditions / Protocol One-step RT-qPCR protocol; supports fast cycling programs with total detection time of 35 minutes or less.
Components / Formulation Premixed one-step RT-qPCR enzyme system (reverse transcriptase plus DNA polymerase).
Reaction buffer containing factors that suppress nonspecific amplification and factors that enhance multiplex amplification efficiency.
dUTP/UDG carryover contamination control system.
