| Product Name | Robust Multiplex qPCR PreMix V1 (UDG Plus, Swab Direct) |
| Catalog No. | RESK-0132 |
| Description | The Robust Multiplex qPCR PreMix V1 (UDG Plus) is a full-premix real-time qPCR reagent built on a new-generation dual-blocking antibody hot-start Taq DNA polymerase. Both the 5'→3' polymerase and 5'→3' exonuclease activities of the enzyme are simultaneously blocked, enabling efficient enzyme blocking and accurate release upon activation. Combined with a uniquely optimized buffer system, an ultra-clean production environment and stringent quality control standards, the reagent delivers further improved amplification performance and stability. Designed for demanding direct-amplification workflows, the PreMix supports direct amplification of oral swab samples when paired with a nucleic acid release reagent, with no loss of performance. The dUTP/UDG carryover prevention system effectively guards against aerosol contamination. |
| Intended Use | For research use in molecular diagnostic assay development: multiplex qPCR detection of DNA pathogen targets, including direct amplification from swab samples and detection of targets such as African swine fever virus (ASFV). |
| Principle / Technology | Dual-blocking antibody hot-start Taq DNA polymerase: antibodies simultaneously block the 5'→3' polymerase and 5'→3' exonuclease activities, ensuring efficient blocking at low temperature and accurate, rapid release of enzyme activity upon heat activation, thereby minimizing nonspecific amplification. Carryover contamination control: the dUTP/UDG system degrades dU-containing carryover PCR products, effectively preventing aerosol contamination. |
| Detection Method | Probe-based real-time quantitative PCR (qPCR); multiplex-capable. |
| Sample Type | DNA templates; swab samples (e.g., throat/oral swabs) processed with a nucleic acid release reagent for direct amplification; extracted pathogen nucleic acids. |
| Performance Range / Specifications | Direct swab amplification: paired with a nucleic acid release reagent, direct amplification performance is unaffected by sample-matrix inhibitors. Excellent full-premix stability: performance stable after 7 days at 37 °C with primers/probes premixed, and after 20 freeze-thaw cycles. Low background residue: no detection in 48-well tests across 11 pathogen targets plus human genomic DNA. High sensitivity: stable detection at Ct 37-39. Fast programs supported: results in 30 minutes. dUTP/UDG anti-contamination system effectively prevents aerosol contamination. Broad platform compatibility: compatible with mainstream real-time qPCR instruments. Excellent amplification efficiency: linear testing of three pathogens over 10 to 10^7 copies/uL gave amplification efficiencies around 96% with R2 >= 0.99. |
| Sensitivity / LOD | Stable detection at Ct 37-39 for low-concentration templates. |
| Specificity | High reagent cleanliness: with primer/probe sets for 11 pathogens plus GAPDH and water as template (48 replicates per target), no nonspecific amplification peaks were observed. Dual-blocking hot-start design minimizes nonspecific amplification and primer-dimer formation. |
| Reaction Conditions / Protocol | Probe-based qPCR; supports fast cycling programs with results in approximately 30 minutes. For swab direct amplification, treat the collected swab sample in a nucleic acid release reagent with vortexing, then add the treated solution directly as template. |
| Components / Formulation | Dual-blocking antibody hot-start Taq DNA polymerase (simultaneous blocking of 5'→3' polymerase and 5'→3' exonuclease activities). Optimized buffer system. dUTP/UDG carryover contamination control system. |
| Storage Conditions | Store at -25 to -15 °C. |
| Shelf Life | 18 months. |
| Package Specifications | 100 T; 1,000 T; 10,000 T |
| Product Form | Liquid full premix. |
| Quality Control | Manufactured in an ultra-clean production environment under stringent quality control standards; background contamination verified by 48-replicate no-template testing across 11 pathogen targets and human genomic DNA with no detection. |
| Expiration Date / Stability | Valid for 18 months when stored at -25 to -15 °C; full premix with primers/probes stable for 7 days at 37 °C and through 20 freeze-thaw cycles. |
| Compatibility | Compatible with mainstream real-time qPCR instruments; suitable for multiple platforms and assay systems. |
| Application Notes / Precautions | For direct swab amplification, use together with a nucleic acid release reagent; vortex the swab sample in the release reagent before adding as template. Successfully applied to the detection of 8 real ASFV clinical samples. The dUTP/UDG anti-contamination system is built in; include a UDG digestion step as required by the protocol. |
