| Product Name | Multiplex Superfast One-Step RT-qPCR Probe Mix (UDG Plus) |
| Catalog No. | RESK-0135 |
| Description | This reagent is designed for multiplex quantitative PCR using RNA as the template. Reverse transcription and quantitative PCR are performed in the same reaction tube, simplifying handling and reducing contamination risk. The kit uses a high-efficiency reverse transcriptase for first-strand cDNA synthesis and a hot-start Taq DNA polymerase for quantitative amplification. It contains an optimized MP Buffer and an Enzymes Mix; the buffer already contains Mg2+ and dNTPs, together with factors that effectively suppress non-specific PCR amplification and enhance multiplex qPCR amplification efficiency. Multiplex amplification is achieved without compromising primer amplification efficiency, and fast cycling programs are supported. A dUTP/UDG system is included to effectively prevent aerosol contamination. |
| Intended Use | For multiplex quantitative detection of RNA targets by one-step RT-qPCR; validated for targets including animal disease viruses and human respiratory viruses, with excellent amplification performance across species. |
| Principle / Technology | One-step RT-qPCR: reverse transcriptase synthesizes first-strand cDNA from the RNA template, followed by probe-based quantitative amplification with hot-start Taq DNA polymerase in the same tube. The dUTP/UDG carryover contamination control system (dUTP replacing dTTP) degrades contaminating amplicons from previous reactions, preventing aerosol-borne contamination and ensuring result accuracy. |
| Detection Method | Probe-based (e.g., TaqMan) multiplex real-time quantitative PCR fluorescence detection. |
| Sample Type | RNA templates (total RNA or purified viral RNA) |
| Performance Range / Specifications | Supports multiplex amplification with high efficiency. Supports superfast programs: a 45-cycle run can be completed in approximately 22 min of actual instrument run time on mainstream real-time PCR instruments. 5x concentrated formulation leaves ample room in the reaction for other components. Broad versatility: validated for amplification of multiple species targets, including animal disease viruses and human respiratory viruses. |
| Sensitivity / LOD | High-sensitivity formulation (superfast, high-sensitivity RT-qPCR mix); quantitative LOD is target- and assay-dependent. |
| Specificity | Optimized buffer contains factors that effectively suppress non-specific PCR amplification while maintaining primer amplification efficiency in multiplex reactions. |
| Reaction Conditions / Protocol | One-tube protocol combining reverse transcription and qPCR. Supports fast cycling programs; a 45-cycle superfast program completes in approximately 22 min of actual run time on mainstream real-time PCR instruments. |
| Components / Formulation | Optimized MP Buffer (containing Mg2+ and dNTPs, with dUTP replacing dTTP, plus factors suppressing non-specific amplification and enhancing multiplex efficiency) Enzymes Mix (reverse transcriptase, hot-start Taq DNA polymerase, and UDG) |
| Storage Conditions | Store at -25 to -15 °C. |
| Shelf Life | 1 year. |
| Package Specifications | 100 T 1,000 T |
| Product Form | Liquid master mix, 5x concentrate (one-tube format) |
| Concentration | 5x concentrate |
| Expiration Date / Stability | Stable for 1 year when stored at -25 to -15 °C. |
| Regulatory / Compliance | For research use only. |
| Compatibility | Compatible with mainstream real-time PCR instruments; validated across a broad range of RNA targets from multiple species. |
| Recommended Buffer System | Optimized MP Buffer containing Mg2+ and dNTPs (supplied in the mix). |
| Application Notes / Precautions | A two-tube product development format is recommended (reagent in one tube, primers/probes in a separate tube). If a combined reagent-plus-primer/probe final product format is required, validation for the specific target is necessary. The dUTP/UDG system effectively prevents aerosol contamination; dUTP replaces dTTP in the reaction. |
