| Product Name | Lyo Multiplex One-Step RT-qPCR Kit (Lyophilizable) |
| Catalog No. | RESK-0089 |
| Description | This kit is a premixed reagent for multiplex quantitative PCR using RNA as template. Reverse transcription and qPCR are performed in the same reaction tube, simplifying the workflow and reducing contamination risk. Its unique buffer and enzyme design makes it suitable for formulating lyophilized one-step RT-qPCR reaction systems and product development. The kit uses a thermostable reverse transcriptase for efficient first-strand cDNA synthesis and a hot-start Taq DNA polymerase for quantitative amplification. It mainly contains an optimized lyophilization buffer and an enzyme mix, supplemented with factors that suppress non-specific PCR amplification and enhance multiplex qPCR amplification efficiency, enabling multiplex fluorescent quantitative amplification while maintaining primer efficiency. A matching lyoprotectant is included, so the reagent can be directly freeze-dried. |
| Intended Use | For one-step, multiplex reverse transcription quantitative PCR (RT-qPCR) detection of RNA targets; suitable for developing lyophilized RT-qPCR products. |
| Principle / Technology | One-step RT-qPCR: thermostable reverse transcriptase synthesizes first-strand cDNA, followed by quantitative amplification with a hot-start Taq DNA polymerase in the same tube. Optimized lyophilization buffer with matched lyoprotectant enables direct freeze-drying. |
| Detection Method | Real-time fluorescence quantitative RT-qPCR (one-step, multiplex). |
| Sample Type | RNA templates (e.g., viral RNA, pseudovirus standards). |
| Performance Range / Specifications | Convenient operation: can be used directly as a liquid reagent or lyophilized for long-term storage Strong reagent stability: after lyophilization, stability remains good following accelerated storage at 50 °C for 25 days Versatile lyophilization formats: supports in-tube (8-strip tube) lyophilization, vial lyophilization, lyophilized beads/microchips and other formats Fast program support: results in as little as 40 min |
| Sensitivity / LOD | Detection sensitivity of the lyophilized beads reaches 5 copies per reaction or better; validated in 4-plex detection of IVA (1,000 copies/mL), IVB (1,000 copies/mL), 2019-nCoV (500 copies/mL) and an internal control (5 uL template input). |
| Specificity | Contains factors that effectively suppress non-specific PCR amplification and improve multiplex amplification efficiency while maintaining primer efficiency. |
| Reaction Conditions / Protocol | Reverse transcription and qPCR are performed in a single tube; compatible with fast programs, delivering results in as little as 40 min. |
| Components / Formulation | 4x Lyo-Buffer (optimized lyophilization buffer) Lyo-Enzyme Mix (thermostable reverse transcriptase and hot-start Taq DNA polymerase) Lyoprotectant |
| Storage Conditions | 4x Lyo-Buffer and lyoprotectant: store at -25 to -15 °C. Lyo-Enzyme Mix: store at 2-8 °C. |
| Shelf Life | 4x Lyo-Buffer and lyoprotectant: 1 year at -25 to -15 °C. Lyo-Enzyme Mix: 6 months at 2-8 °C. |
| Package Specifications | 100 T; 1,000 T; 10,000 T |
| Product Form | Liquid kit components; lyophilizable (supports in-tube lyophilization, vial cakes, lyophilized beads and microchips). |
| Quality Control | Lyophilized products show good morphology: no surface collapse or voids for in-tube lyophilization; beads are plump, round and uniform in size. After accelerated storage at 50 °C for 14 and 25 days, lyophilized beads showed no significant changes in Ct values, fluorescence values or bead morphology when amplifying 10^5, 10^4 and 10^3 copies/mL pseudovirus templates. |
| Expiration Date / Stability | Buffer and lyoprotectant: 1 year at -25 to -15 °C; enzyme mix: 6 months at 2-8 °C. Lyophilized product remains stable after 25 days of accelerated storage at 50 °C. |
| Compatibility | Supports multiple lyophilization formats: in-tube (8-strip) lyophilization, vial lyophilization, lyophilized beads and microchips; suitable for multiplex (4-plex) fluorescent quantitative amplification. |
| Recommended Buffer System | Optimized lyophilization buffer (4x Lyo-Buffer) supplied in the kit. |
| Application Notes / Precautions | Store components at their respective recommended temperatures (buffer/lyoprotectant at -25 to -15 °C; enzyme mix at 2-8 °C). The matched lyoprotectant must be used for freeze-drying. |
