| Product Name | Fast Cells-to-CT One-Step RT-qPCR Probe Mix |
| Catalog No. | RESK-0080 |
| Description | A complete, fast and efficient kit that takes you directly from cells to amplification results, designed for quantitative gene expression analysis of 10 to 10^5 cultured cells (e.g., adherent and suspension cell lines, primary cultured cells, various stem cells and iPS cells). A unique lysis buffer system rapidly releases RNA from cultured cell samples, eliminating the time-consuming RNA purification process; the required RNA template is obtained in only 7 min. Together with the one-step RT-qPCR buffer supplied in the kit, the entire workflow from template preparation to gene expression analysis can be completed in about 1 h. The kit is suitable for high-throughput (96/384-well) analysis of cellular RNA expression. The 100 T format provides 96 lysis reactions, 96 reverse transcription reactions and 96 x 2 qPCR reactions, enough to process one full 96-well cell plate in a single run and avoid contamination and reagent performance loss from repeated opening and freeze-thaw. The 500 T format is intended for 384-well plates and higher-throughput automated screening platforms. |
| Intended Use | Quantitative gene expression analysis directly from cultured cells without RNA purification; suitable for high-throughput 96/384-well cellular RNA expression analysis. |
| Principle / Technology | Direct cell lysis rapidly releases RNA from cultured cells, followed by one-step RT-qPCR in which reverse transcription and probe-based qPCR amplification are performed in a single tube. |
| Detection Method | Probe-based real-time quantitative PCR (fluorescent hydrolysis probe detection) |
| Sample Type | Cultured cells, 10 to 10^5 cells per reaction, including adherent and suspension cell lines, primary cultured cells, various stem cells and iPS cells |
| Performance Range / Specifications | Complete kit covering direct cell lysis and one-step RT-qPCR detection reagents RNA template ready in only 7 min; full workflow from template preparation to gene expression analysis in about 1 h Tolerates up to 30% cell lysate in the reaction Supports up to 384-well gene expression screening in a single run, more than 4x the throughput of conventional methods High precision with universal ROX reference dye included |
| Sensitivity / LOD | Quantitative analysis of gene expression from as few as 10 cultured cells |
| Reaction Conditions / Protocol | 1. Cell lysis: lyse cultured cells with the direct cell lysis reagent; RNA template is released within 7 min without RNA purification 2. One-step RT-qPCR: combine the lysate template with the one-step RT-qPCR buffer, primers and probe; reverse transcription and qPCR amplification are performed in the same tube on a compatible real-time PCR instrument The complete workflow from cells to results takes about 1 h |
| Components / Formulation | Direct cell lysis reagent One-step RT-qPCR detection reagent (one-step RT-qPCR buffer, with universal ROX) |
