| Product Name | DNA Lyo Rapid Isothermal Amplification Kit (Fluorescent, Exo Probe) |
| Catalog No. | RESK-0124 |
| Description | The DNA Lyo Rapid Isothermal Amplification Kit (Fluorescent, Exo Probe) is a molecular biology reagent for rapid, efficient, and specific isothermal amplification of nucleic acid fragments, supplied as an in-situ lyophilized powder. The kit is based on rapid isothermal nucleic acid amplification (RPA) technology: the recombinase UvsX anchors to primers and forms recombinase-primer complexes that scan for homologous target sequences and mediate strand exchange, while single-stranded binding protein stabilizes the displaced single-stranded DNA to form a stable D-loop structure. With the aid of the accessory factor UvsY and a crowding agent, a conformational change in the recombinase-primer complex exposes the primer 3' end, allowing DNA polymerase to bind and initiate extension; upstream and downstream primers amplify simultaneously to generate double-stranded product. Subsequently, with an exonuclease and a target-specific molecular probe designed against the target, the amplification process can be monitored in real time using a fluorescence detection device. The kit enables rapid amplification of target nucleic acids at a constant temperature of 39-42 °C and is widely used in biological research, clinical diagnostics, and molecular testing. |
| Intended Use | Rapid isothermal amplification and real-time fluorescent detection of DNA targets; widely applicable in biological scientific research, clinical diagnostics, and molecular testing. For research use. |
| Principle / Technology | Recombinase polymerase amplification (RPA): recombinase UvsX forms recombinase-primer complexes that mediate strand exchange at homologous target sequences; single-stranded binding protein stabilizes the D-loop; the accessory factor UvsY and a crowding agent facilitate primer 3'-end exposure and DNA polymerase extension from both primers simultaneously. |
| Detection Method | Fluorescent Exo probe method: an exonuclease acts on a target-specific molecular probe during amplification, generating a real-time fluorescence signal monitored on a fluorescence detection device. |
