Product Name C203P1 Multiplex One-Step RT-qPCR Probe Kit (UDG Plus)
Catalog No. RESK-0111
Description This kit performs multiplex quantitative PCR using RNA as the template. Reverse transcription and quantitative PCR are carried out in the same reaction tube, simplifying the workflow and reducing the risk of contamination. A thermostable reverse transcriptase efficiently synthesizes first-strand cDNA, and a hot-start Taq DNA polymerase performs quantitative amplification. The kit includes an optimized MP buffer, an enzyme mix, and a GC enhancer; the buffer already contains Mg2+ and dNTPs, together with factors that suppress non-specific PCR amplification and improve multiplex qPCR amplification efficiency, enabling multiplex amplification while maintaining primer amplification efficiency. A dUTP/UDG system is incorporated to effectively prevent aerosol carryover contamination.
Intended Use One-step multiplex probe-based RT-qPCR detection of nucleic acids from multiple species, including pathogenic microorganisms, human genomic targets, and plants; suitable for research and molecular diagnostic assay development.
Principle / Technology Thermostable reverse transcriptase synthesizes first-strand cDNA from RNA, followed by quantitative amplification with hot-start Taq DNA polymerase and hydrolysis (TaqMan-type) probes in a single closed tube. The dUTP/heat-labile UDG system degrades carryover aerosol contamination to reduce false positives.
Detection Method Real-time fluorescence quantitative PCR (probe-based, multiplex)
Sample Type RNA (total nucleic acid / extracted RNA from pathogens, human, plant, and other species)
Performance Range / Specifications Broad compatibility: one-step RT-qPCR detection of nucleic acids from pathogens, human genomic, plant, and multi-species samples
High amplification sensitivity: optimized buffer system improves detection of low-concentration templates (250 copies/mL)
dUTP/UDG anti-contamination system: efficiently degrades aerosol contamination, reduces false positives
Supports fast protocols: results within 30 min on compatible fast qPCR instruments
Excellent storage stability: stable after 14 days at 37 °C, round-trip shipping, and 10 freeze-thaw cycles
Sensitivity / LOD 250 copies/mL (low-concentration template detection)
Specificity Buffer contains factors that suppress non-specific PCR amplification; dUTP/heat-labile UDG system reduces false positives from carryover contamination.
Reaction Conditions / Protocol One-step protocol: reverse transcription and qPCR in the same tube. Compatible with fast cycling programs (e.g., results within 30 min on a fast 48-well qPCR instrument).
Components / Formulation Optimized MP Buffer (contains Mg2+ and dNTPs)
Enzymes Mix (thermostable reverse transcriptase + hot-start Taq DNA polymerase)
GC-Enhancer
dUTP/heat-labile UDG anti-contamination system
Storage Conditions Store at -25 °C to -15 °C.
Shelf Life 1 year
Package Specifications 100 T; 1000 T; 10000 T
Product Form Liquid master mix kit
Expiration Date / Stability Valid for 1 year at -25 °C to -15 °C. Performance remains stable after 14 days at 37 °C or 4 °C, after round-trip shipping (6 days), and after 10 freeze-thaw cycles.
Compatibility Validated on mainstream real-time PCR instruments (mainstream qPCR platforms); suitable for multiplex assays such as 4-plex respiratory panels and 3-plex veterinary diarrhea panels.
Application Notes / Precautions Use the dUTP/UDG step to control aerosol contamination. Multiplex primer/probe design should be verified for amplification efficiency. Avoid repeated freeze-thaw cycles beyond validated limits.
Batch-to-Batch Consistency Stability-verified manufacturing; performance confirmed stable across shipping, freeze-thaw, and accelerated storage testing.
