| Product Name | 2x TaqMan SNP Genotyping Master Mix |
| Catalog No. | RESK-0094 |
| Description | This product is a 2x concentrated fluorescent quantitative TaqMan SNP genotyping master mix built on a new-generation antibody-based hot-start Taq DNA polymerase. The genetically engineered hot-start Taq polymerase greatly improves amplification sensitivity and specificity, promoting efficient amplification of low-concentration templates. The product is suitable for genotyping and multiplex quantitative gene analysis. The formulation contains a Low ROX reference dye for instruments that require it. |
| Intended Use | SNP genotyping and multiplex quantitative gene analysis by real-time PCR; allelic discrimination assays based on TaqMan probes. |
| Principle / Technology | Allele-specific TaqMan probe genotyping: two differentially fluorescently labeled hydrolysis probes, each perfectly matched to one allele, are combined with flanking PCR primers. During amplification, the 5' exonuclease activity of the hot-start Taq polymerase cleaves only the fully matched probe, generating an allele-specific fluorescent signal; the relative signal intensities of the two reporter dyes discriminate homozygous and heterozygous genotypes. The antibody-based hot-start mechanism suppresses non-specific priming and improves discrimination of low-abundance templates. |
| Detection Method | Real-time quantitative PCR with TaqMan allele-specific hydrolysis probes (endpoint or real-time allelic discrimination) |
| Sample Type | Genomic DNA and other DNA templates (validated on DNA, plasmid DNA, Arabidopsis gDNA and human gDNA); not suitable for RNA templates. Complex lysates such as yeast lysates or viral samples are not recommended. |
| Performance Range / Specifications | Suitable for genotyping and multiplex quantitative gene analysis Amplifies fragments up to <6 kb Stable for one week at room temperature (approx. 23°C) and for 3 months at 4°C Reaction volumes are scalable (e.g., 25 uL reactions by proportional downscaling of all components) |
| Sensitivity / LOD | Genetically engineered hot-start Taq polymerase enables efficient amplification of low-concentration templates, improving detection sensitivity. |
| Specificity | Antibody-based hot-start Taq polymerase delivers high amplification specificity and reliable allele discrimination; the polymerase has no 3'-5' exonuclease (proofreading) activity, and PCR products carry a 3'-A overhang suitable for TA cloning. |
| Reaction Conditions / Protocol | Standard TaqMan probe-based qPCR cycling with allele-specific probes and flanking primers. Reaction volume is flexible; a 25 uL reaction is supported by scaling all components proportionally. Recommended amplicon size: <6 kb. |
| Components / Formulation | 2x master mix containing genetically engineered antibody-based hot-start Taq DNA polymerase, reaction buffer, Mg2+ and dNTPs. Contains a Low ROX passive reference dye. Contains bromophenol blue; free of SDS. |
| Storage Conditions | Store at -25 to -15°C (recommended -20°C). |
| Shelf Life | 2 years when stored at -25 to -15°C. |
| Package Specifications | 1 mL 10 mL 50 mL |
| Product Form | Liquid 2x master mix |
| Concentration | 2x concentrated master mix |
| Activity / Unit Definition | Each 1 mL of master mix contains 50 U of Taq DNA polymerase activity. |
| Expiration Date / Stability | Stable for 2 years at -25 to -15°C; stable for one week at room temperature (approx. 23°C) and for 3 months at 4°C. |
| Regulatory / Compliance | For research use only. |
| Compatibility | Compatible with real-time PCR instruments that require a Low ROX reference dye; suitable for probe-based SNP detection on corresponding platforms. |
| Application Notes / Precautions | The Taq polymerase in this mix has no 3'-5' exonuclease activity; PCR products carry a 3'-A overhang and can be used for TA cloning. Direct restriction digestion of PCR products is not recommended, as some components may interfere with digestion. Direct column purification of PCR products is not recommended, as some components may reduce recovery; gel extraction purification is acceptable. Not recommended for PAGE analysis, as the mix contains components that affect PAGE electrophoresis. Cannot amplify RNA templates. Validated templates include DNA, plasmid DNA, Arabidopsis gDNA and human gDNA; special samples such as yeast lysates or viral material are not recommended. |
For research use only, not for clinical use.
