| Detection Principle | The target antibody is coated in a 96-well microplate to form a solid-phase carrier. Standards or samples are added to the microplates, respectively, and the target is bound to the antibody on the solid-phase carrier. Then, horseradish peroxidase-labeled antibody is added. The unbound antibody is washed off and thoroughly washed again. After that, TMB substrate is added for color development. TMB is converted into blue under the catalysis of peroxidase and then into the final yellow under the action of acid. The depth of the color is positively correlated with the target in the sample. The absorbance (O.D. value) is measured at a wavelength of 450nm using a microplate reader, and the sample concentration is calculated. |
