| Product Name | One-Step Luciferase Assay Kit (High Sensitivity) |
| Catalog No. | RESK-0151 |
| Description | This one-step firefly luciferase reporter gene assay kit is characterized by simple operation and high signal intensity. No cell washing or harvesting is required before the assay; the reagent is added directly to the culture medium, completing cell lysis and detection in a single step and saving experimental time. The kit achieves a relative balance between signal intensity and stability, maintaining relatively high signal intensity while keeping the signal relatively stable for a defined period, ensuring accurate and reliable results. The signal half-life typically exceeds 1 h. The assay has a wide linear range, showing good linearity over a range of up to 200,000 cells per well in a 96-well plate. |
| Intended Use | One-step, high-sensitivity detection of firefly luciferase reporter gene activity in cultured cells, suitable for high-throughput and ultra-high-throughput screening and for functional assays such as ADCC reporter bioassays. |
| Principle / Technology | Firefly luciferase is a protein of approximately 61 kDa. In the presence of ATP, magnesium ions, and oxygen, it catalyzes the oxidation of luciferin to oxyluciferin, emitting bioluminescence at approximately 560 nm. The single reagent formulation lyses cells and supplies the luciferase reaction simultaneously, generating a glow-type signal with a half-life typically exceeding 1 h. |
| Detection Method | Bioluminescence detection using a luminometer or luminescence-capable microplate reader. |
| Sample Type | Cultured cells in medium, including suspension and adherent cells; no medium removal required. Suitable for various cell types and culture conditions. |
| Performance Range / Specifications | Wide linear range: good linearity over up to 200,000 cells per well in a 96-well plate. Signal half-life typically greater than 1 h. Signal intensity essentially unaffected after 5 freeze-thaw cycles. In ADCC functional validation (e.g., against CLDN18.2), delivers high signal, low background, and a clear assay window meeting functional validation requirements. |
| Sensitivity / LOD | Capable of detecting very low concentrations of luciferase, suitable for high-throughput and ultra-high-throughput experimental needs. |
| Reaction Conditions / Protocol | One-step protocol: without washing or harvesting cells, add an equal volume of detection reagent directly to the cell culture medium; cell lysis and detection are completed in one step. Mix briefly and read the luminescence. |
| Components / Formulation | Ready-to-use single detection reagent combining cell lysis and luciferase detection components. |
| Storage Conditions | Store at -80 °C protected from light. If stored at -20 °C protected from light, use within 2 weeks. After opening, aliquot for storage (protected from light). |
| Shelf Life | 1 year for unopened reagent stored at -80 °C protected from light; use within 2 weeks when stored at -20 °C. |
| Package Specifications | 100 T 10 x 100 T 1000 T 10 T |
| Molecular Weight | Firefly luciferase: approximately 61 kDa. |
| Shipping Conditions | Shipped on dry ice. |
| Regulatory / Compliance | For research use only. Not for use in diagnostic procedures. |
| Compatibility | Applicable to various cell types and culture conditions, including suspension and adherent cells; no need to remove culture medium. Suitable for high-throughput and ultra-high-throughput formats. |
| Application Notes / Precautions | No cell washing or collection is required before the assay; add the reagent directly to the culture medium. Protect the reagent from light; after opening, aliquot to avoid repeated freeze-thaw. |
