| Product Name | JC-10 Mitochondrial Membrane Potential Assay Kit |
| Catalog No. | RESK-0198 |
| Description | JC-10 is an upgraded mitochondrial membrane potential fluorescent probe and an improved alternative to JC-1. Although JC-1 is widely used, its poor water solubility causes it to precipitate from aqueous buffers even at 1 μM. JC-10 offers markedly better water solubility and can replace JC-1 in assays requiring higher dye concentrations, although its performance is highly cell-type dependent. In healthy cells, JC-10 selectively accumulates in the mitochondrial matrix, forming reversible red fluorescent aggregates (Ex = 540 nm, Em = 590 nm). In unhealthy mitochondria with decreased or lost membrane potential, JC-10 remains in the cytoplasm in monomeric form, producing green fluorescence (Ex = 490 nm, Em = 525 nm). The color shift allows both qualitative assessment and quantitative ratiometric measurement (red/green fluorescence intensity ratio) of mitochondrial depolarization. The kit includes CCCP as a positive control that induces loss of mitochondrial membrane potential. For samples in six-well plates, the kit is sufficient for 100 assays. |
| Intended Use | For qualitative and quantitative detection of changes in mitochondrial membrane potential in cells by flow cytometry, fluorescence imaging, or fluorescence microplate readers. |
| Principle / Technology | JC-10 is a cationic, ratiometric fluorescent probe. In polarized (healthy) mitochondria it accumulates in the matrix and forms reversible aggregates emitting red fluorescence (Ex = 540 nm, Em = 590 nm); upon mitochondrial depolarization it reverts to monomers in the cytoplasm emitting green fluorescence (Ex = 490 nm, Em = 525 nm). The red/green fluorescence intensity ratio reflects the degree of mitochondrial depolarization. |
| Detection Method | Flow cytometry (green fluorescence in the FL1 channel, red fluorescence in the FL2 channel), fluorescence imaging, or fluorescence microplate detection. |
| Sample Type | Cultured cells (e.g., samples in six-well plates) |
| Performance Range / Specifications | JC-10 has better water solubility than JC-1. Sufficient for 100 assays for samples in six-well plates. Ex/Em: aggregates 540/590 nm; monomers 490/525 nm. |
| Reaction Conditions / Protocol | Use the provided JC-10 Staining Buffer (5X) diluted with ultrapure water for staining; include the provided CCCP-treated sample as a positive control for loss of mitochondrial membrane potential. |
| Components / Formulation | JC-10 fluorescent probe JC-10 Staining Buffer (5X) CCCP (positive control) Ultrapure water |
| Storage Conditions | Store at -20 °C, dry and protected from light; aliquot to avoid repeated freeze-thaw cycles. Ultrapure water and JC-10 Staining Buffer (5X) may also be stored at 4 °C. |
| Shelf Life | 1 year |
| Package Specifications | 100 T |
| Product Form | Kit (liquid components) |
| Shipping Conditions | Shipped on wet ice |
| Expiration Date / Stability | Valid for 1 year when stored at -20 °C protected from light; avoid repeated freeze-thaw cycles. |
| Regulatory / Compliance | For research use only. |
| Compatibility | Compatible with standard flow cytometer filter sets (FL1 and FL2 channels), fluorescence microscopes, and fluorescence microplate readers. |
| Application Notes / Precautions | Protect from light. Aliquot to avoid repeated freeze-thaw cycles. JC-10 performance is highly cell-type dependent; although it outperforms JC-1 in some cell lines, results may vary between cell types. Use the provided CCCP as a positive control for mitochondrial membrane potential loss. |
