Product Name Firefly Luciferase Reporter Gene Assay Kit (Flash Type)
Catalog No. RESK-0148
Description This kit is a flash-type firefly luciferase reporter gene assay kit characterized by high sensitivity for the detection of firefly luciferase expression in mammalian cells. Key features: Stronger lysis capability: thoroughly lyses the vast majority of cell types. Stronger signal: enables accurate detection of weak promoter expression. Higher sensitivity: detects samples containing as few as a few hundred cells. Wider linear range: the linear detection range spans more than 8 orders of magnitude of enzyme concentration.
Intended Use Highly sensitive quantitative detection of firefly luciferase reporter gene expression in mammalian cells for promoter activity analysis, transcriptional regulation studies, and related gene function research.
Principle / Technology Firefly luciferase is a protein of approximately 61 kDa. In the presence of ATP, magnesium ions, and oxygen, it catalyzes the oxidation of luciferin to oxyluciferin, emitting bioluminescence at a wavelength of approximately 560 nm, which can be measured with a luminometer. This kit produces flash-type luminescence kinetics, delivering a high-intensity, transient signal.
Detection Method Bioluminescence detection using a luminometer or a microplate reader equipped with a luminescence module.
Sample Type Cultured mammalian cells / cell lysates.
Performance Range / Specifications Linear detection range spanning more than 8 orders of magnitude of enzyme concentration.
Complete lysis of most cell types.
Accurate detection of weak promoter expression.
Sensitivity / LOD Detects firefly luciferase expression in samples containing as few as a few hundred cells.
Reaction Conditions / Protocol Lyse the cells with the provided lysis buffer, then add the firefly luciferase detection reagent and measure luminescence immediately. As this is a flash-type assay, the signal decays after reagent addition; reading should be completed as quickly as possible, ideally within 5 min. Use of a luminometer with an automatic injector is recommended for plate-based assays. For single-tube manual detection, a 10 s measurement time with a 2 s interval is recommended.
Components / Formulation Cell lysis buffer and firefly luciferase detection reagent.
Storage Conditions Store at -20 °C. The firefly luciferase detection reagent should not be subjected to repeated freeze-thaw cycles; aliquot and store at -20 °C or -80 °C.
Shelf Life 1 year when stored at -20 °C.
Package Specifications 100 T
500 T
1000 T
Molecular Weight Firefly luciferase: approximately 61 kDa.
Shipping Conditions Shipped on dry ice.
Regulatory / Compliance For research use only. Not for use in diagnostic procedures.
Compatibility Compatible with luminometers and microplate readers equipped with a luminescence module.
Application Notes / Precautions Centrifugation after cell lysis is optional.
The firefly signal undergoes decay (quenching) after reagent addition; complete the measurement as soon as possible, preferably within 5 min.
In vitro reporter gene assays require cell lysis to fully release intracellular luciferase, eliminating transfection heterogeneity and uneven substrate penetration and ensuring homogeneous, quantitative signals.
Measurement time settings may be adjusted according to the instrument used; for manual single-tube detection, follow the recommended settings in the manual.

For research use only, not for clinical use.
