| Product Name | One-Step Luciferase Assay Kit (High Brightness) |
| Catalog No. | RESK-0152 |
| Description | This one-step firefly luciferase reporter gene assay kit features simple operation, high sensitivity, and high signal intensity, making it suitable for experiments with stringent sensitivity requirements. No cell washing or harvesting is required before the assay; the reagent is used directly, completing cell lysis and detection in one step, eliminating reagent mixing steps and saving experimental time. |
| Intended Use | One-step, high-sensitivity and high-signal-intensity detection of firefly luciferase reporter gene activity, ideal for experiments with high sensitivity requirements, including samples with low target expression levels. |
| Principle / Technology | Firefly luciferase is a protein of approximately 61 kDa. In the presence of ATP, magnesium ions, and oxygen, it catalyzes the oxidation of luciferin to oxyluciferin, emitting bioluminescence at approximately 560 nm. The single reagent formulation lyses cells and drives a high-intensity luciferase reaction in one step. |
| Detection Method | Bioluminescence detection using a luminometer or luminescence-capable microplate reader. |
| Sample Type | Cultured cells in medium; no cell washing or collection required. |
| Performance Range / Specifications | High signal intensity providing a larger detection window. Signal remains essentially stable within 10 min, with signal variation not exceeding 10%. Good linearity with R2 as high as 0.9978. Signal intensity essentially unaffected after 15 freeze-thaw cycles. |
| Sensitivity / LOD | Detects very low concentrations of luciferase; accurately measures samples with low target expression levels. |
| Reaction Conditions / Protocol | One-step protocol: without washing or harvesting cells, add an equal volume of detection reagent directly to the cell culture medium; cell lysis and detection are completed in one step with no reagent mixing required. Read the luminescence with a luminometer. |
| Components / Formulation | Ready-to-use single detection reagent combining cell lysis and luciferase detection components. |
| Storage Conditions | Store at -80 °C protected from light. If stored at -20 °C protected from light, use within 2 weeks. After opening, aliquot for storage (protected from light). |
| Shelf Life | 1 year for unopened reagent stored at -80 °C protected from light; use within 2 weeks when stored at -20 °C. |
| Package Specifications | 100 T 10 x 100 T 1000 T |
| Molecular Weight | Firefly luciferase: approximately 61 kDa. |
| Shipping Conditions | Shipped on dry ice. |
| Regulatory / Compliance | For research use only. Not for use in diagnostic procedures. |
| Compatibility | Applicable to cultured cells in medium without washing or collection steps. |
| Application Notes / Precautions | The signal remains essentially stable within 10 min (variation not exceeding 10%); readings should preferably be completed within this window. Protect the reagent from light; after opening, aliquot to avoid repeated freeze-thaw (signal essentially unaffected after up to 15 freeze-thaw cycles). |
For research use only, not for clinical use.
