Description Platelet-activating factor (PAF) is a potent phospholipid activator and mediator of many leukocyte functions, platelet aggregation and degranulation, inflammation, and anaphylaxis. It is also involved in changes in vascular permeability, oxidative bursts, leukocyte chemotaxis, and enhanced arachidonic acid metabolism in phagocytes. It is produced by a variety of cells, but particularly those involved in host defense, such as platelets, endothelial cells, neutrophils, monocytes, and macrophages. It is produced continuously by these cells, but in small numbers, and its production is controlled by the activity of PAF acetylhydrolase. Inflammatory cells produce large amounts of PAF in response to specific stimuli.

Detection Principle The kit is a competitive enzyme-linked immunosorbent assay (ELISA). To the microtiter wells pre-coated with mouse platelet-activating factor (PAF) antibody, the sample, standard and HRP-labeled antigen are added sequentially, and then incubated and washed, and then the substrate TMB is used to develop the color, which is converted to blue by peroxidase (HRP), and then converted to the final yellow color under the effect of acid. There is a negative correlation between the color and the amount of mouse platelet-activating factor (PAF) in the sample. The absorbance (OD) is measured at 450 nm using an enzyme meter and the concentration of the sample is calculated.
