Product Name Human Thrombopoietin (TPO) ELISA Kit
Catalog No. PAK-HMM-0011
Description Thrombopoietin (TPO), also known as megakaryocyte growth and development factor (MGDF), is a protein encoded by the TPO gene. Thrombopoietin is a glycoprotein hormone produced by the liver and kidneys that regulates platelet production. It stimulates the production and differentiation of megakaryocytes, which are bone marrow cells that sprout large numbers of platelets. Megakaryopoiesis is the process of cell development that leads to platelet production. The proteins encoded by this gene are humoral growth factors necessary for megakaryocyte proliferation and maturation as well as thrombus formation.
Test Species Human
Synonyms THPO; MGDF; MKCSF; ML; MPLLG; Myeloproliferative Leukemia Virus Oncogene Ligand; Megakaryocyte Growth And Development Factor; Megakaryocyte Stimulating Factor
Applications The kit detects human TPO in samples without significant cross-reactivity with its analogs.
Sample Type Serum, plasma, tissue homogenates, cell lysates, cell culture supernatants and other biological fluids.
Detection Methods Sandwich ELISA
Sensitivity 6.17 pg/mL
Detection Range 15.62-1000 pg/mL
Accuracy The intraplate coefficient of variation is less than 10% and the interplate coefficient of variation is less than 10%.
Storage 2-8°C
Shelf Life 6 months
Detection Principle This kit uses double antibody sandwich method enzyme-linked immunosorbent assay (ELISA). To the microtiter wells pre-coated with human thrombopoietin (TPO) capture antibody, the sample, standard, biotin-labeled detection antibody, HRP enzyme conjugate are added sequentially, with warming and washing in the middle, and then the color is developed with the substrate TMB, which is converted into blue color catalyzed by the enzyme peroxidase (HRP), and then converted into the final yellow color under the action of acid. The shade of color is positively correlated with human thrombopoietin (TPO) in the sample. The absorbance (OD value) is measured at 450 nm with an enzyme meter to calculate the concentration of the sample.
