Product Name Human Thrombopoietin antibodies(TPO-Ab)
Catalog No. PAK-HMM-0012
Description Thrombopoietin (TPO), also known as megakaryocyte growth and development factor (MGDF), is a protein encoded by the TPO gene. Thrombopoietin is a glycoprotein hormone produced by the liver and kidney that regulates platelet production. It stimulates the production and differentiation of megakaryocytes, which are bone marrow cells that can sprout large numbers of platelets. Megakaryopoiesis is the process of cell development that leads to platelet production. The protein encoded by this gene is a humoral growth factor required for the proliferation and maturation of megakaryocytes and for thrombosis. The protein is a ligand for MLP/C_MPL, a product of the myeloproliferative leukemia virus oncogene.
Test Species Human
Synonyms Thrombopoietin antibodies
Applications This kit detects human TPO-Ab in samples without significant cross-reactivity with its analogs.
Sample Type Serum, plasma, tissue homogenates, cell lysates, cell culture supernatants and other biological fluids.
Detection Methods Indirect ELISA
Detection Range Qualitative Testing
Accuracy The intraplate coefficient of variation is less than 10% and the interplate coefficient of variation is less than 10%.
Storage 2-8°C
Shelf Life 6 months
Detection Principle This kit uses an indirect method of enzyme-linked immunosorbent assay (ELISA). To the microtiter wells pre-coated with human thrombopoietin antigen, the sample, positive control, negative control, HRP labeled secondary antibody are added sequentially, with warming and washing in the middle, and the color is developed with the substrate TMB, which is converted to blue color catalyzed by peroxidase (HRP), and converted to the final yellow color in the presence of acid. There is a positive correlation between the color and the human thrombopoietin antibody (TPO-Ab) in the sample. The absorbance (OD value) is measured at 450nm using an enzyme meter to determine the negative and positive of the sample.
