Product Name ELISA Kit for Thrombopoietin antibodies (TPO-Ab)
Catalog No. PAK-HMM-0042
Description Thrombopoietin (TPO), also known as megakaryocyte growth and development factor (MGDF), is a protein encoded by the TPO gene. Thrombopoietin is a glycoprotein hormone produced by the liver and kidney that regulates platelet production. It stimulates the production and differentiation of megakaryocytes, which are bone marrow cells that can sprout large numbers of platelets. Megakaryopoiesis is the cellular developmental process that leads to platelet production. The protein encoded by this gene is a humoral growth factor required for the proliferation and maturation of megakaryocytes and for thrombosis. The protein is a ligand for MLP/C_MPL, a product of the myeloproliferative leukemia virus oncogene.
Test Species Mouse
Synonyms Thrombopoietin antibodies; TPOAb
Applications This kit detects TPO-Ab in mice in samples without significant cross-reactivity with its analogs.
Sample Type Serum, plasma, tissue homogenates, cell lysates, cell culture supernatants and other biological fluids.
Detection Methods Indirect ELISA
Detection Range Qualitative Testing
Accuracy The intraplate coefficient of variation is less than 10% and the interplate coefficient of variation is less than 10%.
Storage 2-8°C
Shelf Life 6 months
Detection Principle This kit is an indirect enzyme-linked immunosorbent assay (ELISA). In this kit, the sample, positive control, negative control and HRP-labeled secondary antibody are added sequentially into the wells pre-coated with mouse plasminogen antigen, and then incubated and washed, and then the substrate TMB is used to develop the color, which is converted into blue color by peroxidase (HRP), and then converted into yellow color under the action of acid. There is a positive correlation between the color and the presence of mouse thrombopoietin antibody (TPO-Ab) in the sample. The absorbance (OD) is measured at 450nm to determine the negativity of the sample.
