| Product Name | Cell Senescence beta-Galactosidase Staining Kit |
| Catalog No. | RESK-0147 |
| Description | Cell senescence is a safeguard mechanism by which cells control their growth potential, generally referring to replicative senescence (RS): after a limited number of divisions, normal cells stop dividing and, although still viable, undergo marked changes in morphology and physiological/metabolic activity. Commonly used biological hallmarks of senescent cells in vitro include: 1) irreversible growth arrest; and 2) activation of senescence-associated beta-galactosidase (SA-beta-gal). As a lysosomal hydrolase, beta-galactosidase normally shows activity at pH 4.0, but in senescent cells it is active at pH 6.0, and the number of SA-beta-gal-positive cells in a population increases with passage number. This kit is designed based on the change in SA-beta-gal activity: using X-Gal as the substrate, a deep blue product is generated under catalysis by senescence-specific beta-galactosidase, allowing the senescence status of cells to be analyzed by color change under a light microscope. |
| Intended Use | Detection of cellular senescence in cultured cells and in frozen tissue sections by staining of senescence-associated beta-galactosidase (SA-beta-gal) activity. For research use only. |
| Principle / Technology | Based on senescence-associated beta-galactosidase (SA-beta-gal) activity at pH 6.0. X-Gal is hydrolyzed by senescence-specific beta-galactosidase to produce an insoluble deep blue product, which is visualized by light microscopy. The kit stains only senescent cells and does not stain presenescent cells, quiescent cells, immortal cells or tumor cells. |
| Detection Method | Colorimetric (chromogenic) detection; deep blue precipitate observed by light microscopy. |
| Sample Type | Cultured cells and frozen tissue sections. Not recommended for paraffin-embedded sections, because temperature and fixatives during paraffin embedding may inactivate beta-galactosidase and cause staining failure. |
| Performance Range / Specifications | 100 tests. Stable performance with optimized parameters, sensitive color development and high specificity. |
| Specificity | Stains only senescent cells; does not stain presenescent cells, quiescent cells, immortal cells or tumor cells. |
| Reaction Conditions / Protocol | 1. Thaw the beta-galactosidase staining solution completely; if precipitate is present, it must be fully dissolved before use. 2. Fix the cells or frozen sections and wash. 3. Add the X-Gal staining working solution and incubate at pH 6.0 (do not perform the staining reaction in a CO2 cell culture incubator, because the high CO2 concentration alters the pH of the working solution and causes staining failure). 4. Observe the deep blue color by light microscopy to assess senescence. |
| Components / Formulation | Kit containing X-Gal solution and beta-galactosidase staining solutions/buffers (per kit configuration). |
| Storage Conditions | Store at -20 °C; the X-Gal solution must be stored protected from light. |
| Shelf Life | 1 year when stored at -20 °C. |
| Package Specifications | 100 tests |
| Product Form | Liquid kit components. |
| pH Range / Optimal pH | Staining is performed at pH 6.0, the pH at which senescence-associated beta-galactosidase is active (ordinary lysosomal beta-galactosidase is active at pH 4.0). |
| Shipping Conditions | Shipped with ice packs. |
| Expiration Date / Stability | 1 year at -20 °C; keep the X-Gal solution protected from light. Frozen sections stored for extended periods may show degraded beta-galactosidase and weak staining. |
| Application Notes / Precautions | Not recommended for paraffin-embedded sections (embedding temperature and fixatives may inactivate beta-galactosidase). Do not perform the staining reaction in a CO2 incubator; high CO2 alters the pH of the staining working solution and causes staining failure. If color development is weak or absent, possible causes include: no successful senescence induction in the sample, incompletely dissolved reagent (dissolve any precipitate fully after thawing), or prolonged storage of frozen sections leading to enzyme degradation. Protect the X-Gal solution from light. |
For research use only, not for clinical use.
