Description A porcine-derived trypsin-EDTA solution at 0.25% trypsin with 0.02% EDTA for the enzymatic detachment of adherent mammalian cells from culture vessel surfaces during routine subculturing.
Intended Use Dissociation of adherent mammalian cell monolayers for passaging, cell counting, and preparation of single-cell suspensions for downstream applications.
Principle / Technology Trypsin cleaves peptide bonds at the carboxyl side of lysine and arginine residues, digesting extracellular matrix and cell adhesion proteins; EDTA chelates calcium and magnesium ions, disrupting integrin-mediated cell-substrate adhesion at focal adhesions.

Reaction Conditions / Protocol Wash monolayer with PBS without Ca2+ and Mg2+; add trypsin-EDTA to cover cell surface (1–2 mL per T-75 flask); incubate at 37°C until cells round up (2–5 min); neutralize with complete medium containing FBS; centrifuge and resuspend.
