| Product Name | Topoisomerase I (20 U/uL) |
| Catalog No. | IVDRM-0515 |
| Description | A type I topoisomerase obtained from vaccinia virus by genetic recombination, Topoisomerase I relaxes the supercoiled structure of DNA and also recognises and cuts duplex DNA ends carrying the 5 prime C(T)CCTT motif, forming a stable covalent complex with the DNA. That complex can rejoin the 5 prime hydroxyl of a DNA molecule to restore an intact strand, a property used to prepare TOPO cloning vectors where it supports positive clone rates up to 100%, and to join adapters during next generation sequencing library construction. The preparation is free of exonuclease residue and carries E. coli genomic DNA below 1 copy per 5 U. |
| Intended Use | TOPO cloning vector preparation and adapter ligation in NGS library construction |
| Principle / Technology | Relaxation of supercoiled DNA and covalent complex mediated joining of the 5 prime C(T)CCTT motif to a 5 prime hydroxyl end |
