| Product Name | Thrombin from Bovine Plasma, High Specific Activity (above 2000 IU/mg) |
| Catalog No. | IVDRM-0296 |
| Description | Thrombin is best known for turning fibrinogen into a clot, and in that role it releases the A and B peptides so monomers polymerise into a fibrin network; it is used diagnostically in clotting assays, factor measurements and defibrination of blood or plasma. The same precision makes it a useful tool protease: it cuts only after a specific short motif, so a recognition site engineered into a fusion protein gives single-site cleavage with high efficiency. This preparation comes from bovine plasma, is specified above 2000 IU per milligram of protein and shows no contaminating protein bands by SDS-PAGE, with moisture no higher than 5 percent. Two chains of 31 kDa and 6 kDa are held together by a disulphide bond and make up the 37 kDa molecule. |
| Intended Use | Coagulation assays, clotting factor determination, defibrination of blood or plasma, and site-specific cleavage of recombinant fusion proteins carrying a thrombin recognition sequence. |
| Principle / Technology | As a serine protease it hydrolyses the bond following arginine within an Arg-containing recognition sequence, converting fibrinogen to fibrin in coagulation and cleaving engineered linkers on the same chemistry. |
| Sample Type | Plasma, whole blood for defibrination, and purified fusion proteins in solution. |
| Performance Range / Specifications | Specific activity above 2000 IU per milligram of protein; no contaminating protein bands by SDS-PAGE; no other protease activity; effective virus inactivation and consistent production. |
| Sensitivity / LOD | Does not apply; sensitivity comes from the downstream detection chemistry. |
| Specificity | Molecular weight 37 kDa composed of 31 kDa and 6 kDa chains; CAS 9002-04-4; specific activity above 2000 IU per milligram of protein; moisture not above 5 percent weight by weight; supplied as 1 KU or 2 KU of lyophilised powder. |
| Reaction Conditions / Protocol | Reconstitute the powder in water or a mild buffer, add it to the reaction at the calculated activity ratio, incubate under the chosen conditions, then stop the digestion or remove the enzyme according to the downstream need. |
| Components / Formulation | One vial of lyophilised thrombin powder per order. |
| Storage Conditions | Store at 2 to 8 °C. |
| Shelf Life | 3 years from receipt when refrigerated. |
| Package Specifications | Supplied as 1 KU or 2 KU of lyophilised powder. |
| Product Form | Lyophilised powder. |
| Quality Control | Released on specific activity, absence of contaminating bands by SDS-PAGE and moisture content. |
| Key Features | - Specific activity above 2000 IU per milligram of protein - Prefers A-B-Pro-Arg at the cleavage point with Leu-Val-Pro-Arg-Gly-Ser and Gly-Arg-Gly as common recognition sequences - No contaminating protein bands by SDS-PAGE - Moisture kept at or below 5 percent weight by weight - Three-year refrigerated shelf life |
| Purity | SDS-PAGE shows no contaminating protein bands. |

| Activity / Unit Definition | Expressed as international units per milligram of protein. |
| Molecular Weight | 37 kDa overall, made of 31 kDa and 6 kDa chains joined by a disulphide bridge. |
| Source / Origin | Purified from bovine plasma. |
| pH Range / Optimal pH | Not critical across the working range; reaction pH is set by the chosen assay. |
| Shipping Conditions | Shipped cold. |
| Expiration Date / Stability | Remains usable for the full shelf life provided the temperature recommendation is followed. |
| Regulatory / Compliance | For research use only. |
| Compatibility | Appropriate for both coagulation-based assays and tag cleavage in molecular biology workflows. |
| Recommended Buffer System | Reconstitute in water or a buffer chosen by the downstream protocol. |
| Application Notes / Precautions | This material is of animal origin, so it is unsuitable where an animal-free declaration is required; note also that it is highly active against fibrinogen, which may interfere in clotting-sensitive applications. |
| Batch-to-Batch Consistency | Lot-to-lot consistency is maintained through defined manufacturing specifications; every lot ships with its release record. |
