| Product Name | Thermostable RNase T1 (500 U/uL) |
| Catalog No. | IVDRM-0491 |
| Description | A thermostable variant obtained from RNase T1 by directed protein evolution, this ribonuclease keeps 100% of its activity at 50°C, whereas the wild type enzyme loses at least half of its activity at the same temperature. Raising the reaction temperature helps fully open RNA secondary structure and limits its interference with digestion, so the enzyme delivers more complete cleavage. Like the parent enzyme, it cuts single stranded RNA specifically after guanine ribonucleotides to leave 3 prime phosphate ends. Supplied at 500 U/uL, one unit is defined as the amount of enzyme that produces a change in A260 of 1.0 in 37°C pH 7.5 buffer with yeast RNA as substrate. |
| Intended Use | Thorough cleavage of RNA for secondary structure analysis and complete digestion studies |
| Principle / Technology | Guanyl specific cleavage of single stranded RNA at elevated temperature via a 2 prime, 3 prime cyclic phosphate intermediate |
| Sample Type | Single stranded RNA |
| Specificity | Cuts specifically after guanine ribonucleotides; retains 100% activity at 50°C where the wild type loses at least 50% |
| Reaction Conditions / Protocol | Incubate with the RNA substrate at elevated temperature in pH 7.5 buffer; one unit gives an A260 change of 1.0 with yeast RNA |
| Storage Conditions | -25 to -15°C |
| Shelf Life | 2 years |
| Package Specifications | 100 KU; 5 x 100 KU |
| Key Features | Directed evolution variant; full activity maintained at 50°C for more complete digestion |
