Product Name Taq DNA Ligase for Ligase Chain Reaction and Allele-Specific Detection
Catalog No. IVDRM-0500
Description A thermostable DNA ligase that joins the 5 prime phosphate and 3 prime hydroxyl ends of two oligonucleotides hybridised in adjacent positions on the same complementary target strand. Ligation occurs only when both oligonucleotides are perfectly paired with the target and leave no gap between them, which makes single base substitutions directly detectable through ligase chain reaction and ligase detection reaction formats. The enzyme uses NAD+ as cofactor and stays active across 45 to 65°C, and it also supports introduction of mutations through incorporation of phosphorylated oligonucleotides during primer extension amplification.
Intended Use Allele-specific detection by ligase chain reaction and ligase detection reaction, single nucleotide variation analysis and mutation introduction
Principle / Technology NAD-dependent ligation of adjacent perfectly paired oligonucleotides on a complementary target strand between 45 and 65°C
Sample Type Oligonucleotide probes hybridised to a complementary target DNA strand
Specificity Ligates only perfectly paired adjacent oligonucleotides with no gap, enabling single base discrimination
Reaction Conditions / Protocol Incubate with paired probe oligonucleotides and target DNA at 45 to 65°C in an NAD+ containing buffer
Storage Conditions -20°C, avoid repeated freeze thaw; store the B component buffer at -80°C for periods beyond 30 days
Shelf Life 1 year
Package Specifications 1000 U; 2000 U; 10000 U
Key Features Thermostable across 45 to 65°C; gap-free perfect pairing requirement enables single base discrimination
