| Product Name | T7 Endonuclease I (10 U/uL) |
| Catalog No. | IVDRM-0475 |
| Description | Derived from bacteriophage T7, this nuclease recognises and cuts incompletely paired DNA, cruciform structures, Holliday junctions, crossed DNA and heteroduplexes, and also nicks double stranded DNA at a slower rate. Cleavage falls on the first, second or third phosphodiester bond on the 5 prime side of the mismatch. After genome editing, insertions, deletions or point mutations create mismatched duplexes at the target locus, and the enzyme cuts near these sites to release fragments of defined length that report editing efficiency. |
| Intended Use | Assessment of genome editing efficiency through mismatch cleavage |
| Principle / Technology | Recognition and cleavage of mismatched and branched DNA structures on the 5 prime side of the mismatch |
| Sample Type | Heteroduplex DNA from edited samples |
| Specificity | Mismatched DNA, cruciform DNA, Holliday junctions, crossed DNA and heteroduplexes |
| Reaction Conditions / Protocol | Incubate the heteroduplex sample with the enzyme and resolve the cleavage products; 20 U cleaves 200 ng of mismatched double stranded DNA efficiently |
| Storage Conditions | -25 to -15°C |
| Shelf Life | 2 years |
| Package Specifications | 250 U; 2500 U |
