| Product Name | T4 DNA Polymerase, 3′→5′ Exonuclease-Deficient Mutant (5 U/uL) |
| Catalog No. | IVDRM-0507 |
| Description | T4 DNA polymerase, also known as T4 gp43, is a polymerase from bacteriophage T4 that synthesises DNA in the 5 prime to 3 prime direction in the presence of template and primer. This variant carries the D219A point mutation, which greatly reduces the 3 prime to 5 prime exonuclease activity while leaving the polymerase activity fully intact, so it is suited to seamless cloning workflows where controlled processing of DNA ends is required without strong backward exonuclease digestion. |
| Intended Use | Seamless cloning of DNA fragments |
| Principle / Technology | 5 prime to 3 prime DNA synthesis from template and primer with strongly reduced 3 prime to 5 prime exonuclease digestion |
| Sample Type | DNA fragments for seamless cloning |
| Specificity | D219A point mutation greatly lowers 3 prime to 5 prime exonuclease activity with full polymerase activity retained |
| Reaction Conditions / Protocol | Incubate with the DNA substrate under reaction conditions appropriate to the cloning workflow |
| Storage Conditions | -25 to -15°C |
| Shelf Life | 2 years |
| Package Specifications | 200 U; 1000 U |
| Key Features | Exonuclease-deficient mutant retains complete polymerase activity; suited to seamless cloning |
| Concentration | 5 U/uL |
| Readouts / Downstream Assays | Agarose gel electrophoresis of processed fragments |
For in vitro diagnostic use.
