Product Name Syringe Filters, PVDF Membrane, 0.22 um Pore Size, 13 mm Diameter, Sterile, Non-Pyrogenic
Catalog No. IGM-HMM-0072
Description Sterile syringe filters with hydrophilic polyvinylidene fluoride (PVDF) membrane, 0.22 um pore size, and 13 mm diameter housing, individually blister-packed and gamma-irradiated. The hydrophilic PVDF membrane provides low protein binding (typically <5 ug/cm^2 BSA binding), very low extractables, and broad chemical compatibility, making it the filter of choice for sterilizing aqueous solutions, tissue culture media, buffers, and biological fluids. The 0.22 um pore size reliably retains bacteria, fungi, and particulates while allowing efficient flow of aqueous solutions. The 13 mm diameter is optimized for small volume filtration (1-10 mL) with standard Luer-lock or Luer-slip syringes. Female Luer-lock inlet and male Luer-slip outlet ensure secure, leak-free connections.
Intended Use Sterile filtration of: aqueous solutions (buffers, media, reagents); protein and antibody solutions; cell culture media and supplements; pharmaceutical and biotechnology solutions; HPLC and UPLC mobile phases (aqueous); clarification of small-volume samples prior to chromatography; and removal of bacterial contaminants from heat-sensitive biological fluids.
Principle / Technology Size-exclusion filtration: the PVDF membrane contains a network of interconnected pores with 0.22 um nominal pore size rating. Particles larger than the pore size (including bacteria: 0.2-2 um; yeast: 3-5 um; particulates: >0.22 um) are retained on the membrane surface or within the depth of the membrane matrix. Hydrophilic PVDF is naturally water-wettable without pre-wetting, enabling direct filtration of aqueous solutions without surfactant treatment. Low protein binding minimizes adsorptive losses of dilute protein solutions.
Detection Method 1) Aseptically remove filter from blister pack; 2) Attach Luer-lock syringe (filled with solution to be filtered) to filter inlet (female Luer-lock); 3) Attach sterile receiving vessel or needle to filter outlet (male Luer-slip); 4) Apply gentle, steady pressure to syringe plunger; filter into sterile container; 5) For maximum recovery, expel residual volume with a small volume of air after filtration.
Sample Type Aqueous solutions: buffers, cell culture media, serum-containing media, protein solutions, antibody solutions, DNA/RNA solutions, pharmaceutical preparations, and HPLC mobile phases; sample volume 1-10 mL per filter with standard syringe (filter capacity 5-20 mL depending on solution particulate load).
Performance Range / Specifications Membrane: hydrophilic PVDF; pore size: 0.22 um (nominal); diameter: 13 mm; effective filtration area: approximately 0.65 cm^2; housing: polypropylene; connections: female Luer-lock inlet, male Luer-slip outlet; hold-up volume: <30 uL (post-air purge); burst pressure: >60 psi (4.1 bar); flow rate: >15 mL/min for water at 10 psi (gravity to moderate pressure); sterility: SAL 10^-6 (gamma irradiated).
Sensitivity / LOD Bacterial retention: retains >10^7 CFU Brevundimonas diminuta per cm^2 (ASTM F838 standard); particle retention: >99.99% of 0.22 um and larger particles.
Specificity PVDF membrane: hydrophilic, low protein binding; compatible with aqueous and mild organic solutions (alcohols up to 50%, DMSO up to 50%, acetonitrile up to 50%); not recommended for strong acids (concentrated HNO3, H2SO4, HCl), strong bases (NaOH >1 M), or halogenated solvents (chloroform, dichloromethane).
Reaction Conditions / Protocol Filtration at RT; flow rate decreases as membrane becomes loaded with particulates; replace filter if flow rate drops significantly or if >20 mL filtered through single 13 mm filter.

Compatibility Compatible aqueous solutions: water, buffers (phosphate, Tris, HEPES, carbonate, borate, acetate, citrate), saline, cell culture media (DMEM, RPMI, MEM, DMEM/F12, IMDM) with or without serum (0-20%), protein solutions, antibody solutions, nucleic acid solutions, sugar solutions, and most biological fluids. Limited organic solvent compatibility: alcohols (methanol, ethanol, isopropanol up to 50%), DMSO (up to 50%), acetonitrile (up to 50%), acetone (up to 30%). Not compatible: concentrated mineral acids (HCl >1 M, HNO3 >1 M, H2SO4 >1 M), strong bases (NaOH >1 M), halogenated solvents (dichloromethane, chloroform), THF, DMF, and pyridine. For DMSO >50% or aggressive organic solvents, use PTFE membrane filters.
Recommended Buffer System Compatible with all standard aqueous buffer systems; low extractables compatible with HPLC and LC-MS mobile phases.
Application Notes / Precautions Pre-wet the filter by passing 0.5-1 mL of solution through the filter and discarding the filtrate (removes any potential extractables from the membrane and housing). For protein solutions: PVDF has the lowest protein binding among common membrane types (PVDF < PES < nylon < nitrocellulose); use PVDF for dilute protein solutions to maximize recovery. When filtering cell culture media with serum: serum proteins may coat the membrane and reduce effective pore size — use a 0.45 um pre-filter for serum-containing media followed by 0.22 um sterilization. Apply gentle, steady pressure; excessive pressure may cause membrane rupture or housing failure. Do not exceed 60 psi. For maximum recovery of small volumes, purge filter with a small volume of air after the solution has passed through. Single-use only; do not reuse or resterilize.
Batch-to-Batch Consistency Bubble point >16 psi for every lot; bacterial retention LRV >7; flow rate >15 mL/min; protein binding <5 ug/cm^2; sterile and non-pyrogenic.
