Product Name ssRNA CircleLigase for Circular RNA Synthesis (70 uM)
Catalog No. IVDRM-0581
Description A recombinant single-stranded RNA adenylyl cyclase expressed in E. coli, ssRNA CircleLigase catalyses both intramolecular circularisation and intermolecular joining of ssRNA. Against 18 nucleotide ssRNA substrates it keeps high cyclisation activity above 60°C and works best between 85 and 90°C, a thermostability beyond available T4 RNA Ligase 1 and CircLigase. The enzyme depends on ATP, with concentrations below about 100 uM favouring product and higher concentrations increasing concatemers and adenylated AppRNA intermediates, while more than 500 uM ATP markedly suppresses overall activity. Main uses are efficient in vitro circular RNA synthesis, construction of exonuclease-resistant aptamers and rolling circle amplification templates for molecular diagnostics.
Intended Use In vitro circular RNA synthesis, exonuclease-resistant aptamer construction and rolling circle amplification template preparation
Principle / Technology ATP-dependent intramolecular circularisation and intermolecular joining of single-stranded RNA
Sample Type Short single-stranded RNA substrates such as 18 nucleotide oligoribonucleotides
Specificity Optimal reaction at 85 to 90°C; low ATP favours adenylation while more than 500 uM ATP inhibits activity
Reaction Conditions / Protocol Incubate at 85 to 90°C for 1 hour with a controlled ATP concentration and stop with 2 mM or more EDTA
Storage Conditions -25 to -15°C
Shelf Life 2 years
Package Specifications 2000 pmol
Key Features High temperature cyclisation above 60°C; a defined ATP window favours circular product over concatemers
Purity Greater than or equal to 95%
Concentration 70 uM
Recommended Buffer System Tris-HCl, KCl, DTT and EDTA with 50% glycerol at pH 7.4
Readouts / Downstream Assays Circular versus linear RNA resolved by denaturing gel electrophoresis

For in vitro diagnostic use.
