Product Name SgeI Restriction Endonuclease, Methylation-Dependent (5 U/uL)
Catalog No. IVDRM-0532
Description Unlike ordinary restriction enzymes, SgeI cuts DNA targets that contain 5-methylcytosine on single or double stranded DNA, recognising the m5CNNG(N)9/(N)13 site and cleaving best at 37°C in its dedicated buffer. The enzyme always cuts DNA methylated by the Dcm methyltransferase and also cuts targets overlapping CpG methylation, which makes it valuable for workflows that read out cytosine methylation state. To keep performance consistent, the storage buffer carries pre-mixed BSA, which stabilises the enzyme and binds contaminants that may be present in DNA preparations.
Intended Use Methylation state dependent cleavage of cytosine methylated DNA targets
Principle / Technology Recognition of the m5CNNG(N)9/(N)13 site with cleavage dependent on 5-methylcytosine in the target
Sample Type Dcm or CpG methylated double stranded DNA
Specificity Always cuts Dcm methylated DNA; cuts targets overlapping CpG methylation; inactive on unmethylated targets
Reaction Conditions / Protocol Digest in 1x dedicated buffer at 37°C; heat inactivate at 80°C for 20 minutes
Storage Conditions -25 to -15°C
Shelf Life 2 years
Package Specifications 250 U
Key Features Methylation dependent specificity; storage buffer with pre-mixed BSA for consistent performance
Concentration 5 U/uL
Readouts / Downstream Assays Gel readout distinguishing methylated and unmethylated substrates

For in vitro diagnostic use.
