Product Name RNase HII, Glycerol-Free (2 U/uL)
Catalog No. IVDRM-0489
Description A ribonucleoside endonuclease from Pyrococcus abyssi produced by recombinant expression in E. coli, RNase HII recognises DNA-rN-DNA/DNA duplexes and cuts at the site where a ribonucleotide has been incorporated into DNA, while showing very low activity on single stranded RNA and none on single or double stranded DNA. Cleavage occurs from the 5 prime side of the ribonucleotide to leave a 5 prime phosphate and a 3 prime hydroxyl group. Activity is optimal between 70 and 75°C across a 50 to 75°C working range and is minimal at room temperature, and the enzyme tolerates 45 minutes at 95°C with almost no loss of activity. The glycerol-free formulation, stable at 2 to 8°C, suits lyophilised reaction systems.
Intended Use LAMP probe assays, RNase HII dependent PCR and studies of ribonucleotide excision during DNA replication and repair
Principle / Technology Endonucleolytic cleavage at ribonucleotide sites within DNA-rN-DNA/DNA duplexes, releasing 5 prime phosphate and 3 prime hydroxyl ends
