Product Name RNase HII (2 U/uL)
Catalog No. IVDRM-0498
Description A ribonucleoside endonuclease from Pyrococcus abyssi produced by recombinant expression in E. coli, RNase HII recognises DNA-rN-DNA/DNA duplexes and cuts at the site where a ribonucleotide is incorporated into DNA, while showing very low activity on single stranded RNA and none on single or double stranded DNA. Cleavage occurs from the 5 prime side of the ribonucleotide residue and leaves a 5 prime phosphate and a 3 prime hydroxyl group. Activity peaks between 70 and 75°C across a 50 to 75°C working range and is minimal at room temperature, and the enzyme tolerates 45 minutes at 95°C with almost no loss of activity, so it is compatible with PCR reaction systems.
Intended Use LAMP probe assays, RNase HII dependent PCR, excision of misincorporated ribonucleotides and degradation of Okazaki fragment RNA
Principle / Technology Endonucleolytic cleavage at ribonucleotide sites within DNA-rN-DNA/DNA duplexes, releasing 5 prime phosphate and 3 prime hydroxyl ends
Sample Type DNA-rN-DNA/DNA duplexes; LAMP and rhPCR reaction mixes
Reaction Conditions / Protocol Add to the reaction mix; activity is optimal at 70 to 75°C and the enzyme tolerates 95°C for 45 minutes
