Product Name Red Nucleic Acid Gel Stain (10,000× in Water)
Catalog No. IVDRM-0106
Description Red Nucleic Acid Gel Stain is a unique oily macromolecule that cannot penetrate cell membranes and is not volatile or sublimable, so it cannot be inhaled. Ames test results demonstrate that the stain is completely non-mutagenic at gel-staining concentrations, making it a safe, non-toxic nucleic acid dye. It shares the same spectral characteristics as ethidium bromide (EB), so no change of filters or observation equipment is required—it can be detected on a standard UV gel transilluminator with excitation at 300 nm. The stain is suitable for staining dsDNA, ssDNA, and RNA in agarose and polyacrylamide gel electrophoresis, and can be applied either by in-gel (pre-cast) staining or post-electrophoresis soaking, offering convenient and flexible operation. Spent stain decomposes naturally in the environment and requires no special disposal.
Intended Use Staining of dsDNA, ssDNA, and RNA of all fragment sizes in agarose and polyacrylamide gel electrophoresis, either before (in-gel) or after (post-staining) electrophoresis.
Principle / Technology A safe, non-mutagenic fluorescent nucleic acid dye and EB alternative; it binds nucleic acids through electrostatic and charge interactions and fluoresces when bound, with spectral characteristics identical to EB.
Detection Method UV excitation at 300 nm on a standard ultraviolet gel transilluminator (same filters and observation setup as for EB); emission wavelength 590 nm.
Sample Type dsDNA, ssDNA, and RNA of various fragment sizes separated in agarose or polyacrylamide gels.
Performance Range / Specifications Supplied at 10,000× concentration in water
Excitation wavelength: 300 nm; emission wavelength: 590 nm
Sensitivity comparable to EB
Stable at room temperature with a 5-year shelf life
Suitable for staining agarose or polyacrylamide gels before or after electrophoresis
Post-staining solution can be reused approximately 3 times
Sensitivity / LOD Sensitivity equivalent to ethidium bromide, enabling efficient electrophoresis staining.
Specificity Binds nucleic acids (dsDNA, ssDNA, RNA) via electrostatic and charge interactions.
Reaction Conditions / Protocol In-gel staining: add the 10,000× stain to the molten gel solution before casting. Post-staining: soak the gel in diluted staining solution after electrophoresis. Tip for easy mixing: after boiling the gel solution, pipette the stain into a clean empty flask while pouring in the gel solution so the stream disperses the dye evenly.
Components / Formulation Red Nucleic Acid Gel Stain (10,000× in Water)
Storage Conditions Store at room temperature protected from light. Do not freeze at -20°C; freezing may damage the dye and cause precipitation.
Shelf Life 5 years.
Package Specifications 500 μL
Product Form Liquid (10,000× aqueous solution)
Quality Control Ames test confirms the stain is non-mutagenic at gel-staining concentrations.
Concentration 10,000× in water
Expiration Date / Stability Stable at room temperature for 5 years when protected from light.
Regulatory / Compliance Safe and non-toxic: completely non-mutagenic at gel-staining concentrations (Ames test), offering a safe alternative to ethidium bromide. The water-based formulation avoids the potential hazards of DMSO, which can be absorbed through skin. Waste stain decomposes naturally in the environment without special disposal.
Compatibility Compatible with downstream applications including gel recovery, cloning, and sequencing. Identical spectral properties to EB, so existing UV transilluminators, filters, and imaging setups can be used without modification.
Application Notes / Precautions The aqueous formulation performs equivalently to DMSO-based versions in agarose gel electrophoresis while avoiding the skin-absorption hazards of DMSO.
Post-staining solution can be reused approximately 3 times.
Because the dye binds nucleic acids tightly via electrostatic interaction, high sample loading may affect nucleic acid mobility and cause smearing or band distortion; if this occurs, reduce the nucleic acid loading amount and dilute the marker.
Do not store at -20°C. If accidentally frozen for a short period, thaw and warm at about 35°C, sonicate for 1-2 min before use, and avoid further freeze-thaw cycles.
For easy mixing, add the dye while pouring the boiled gel solution into a clean empty flask so the gel stream disperses the dye.
