Product Name Red Nucleic Acid Gel Stain (10,000× in DMSO)
Catalog No. IVDRM-0109
Description Red Nucleic Acid Gel Stain (10,000× in DMSO) from Alta DiagnoTech is a safe, non-toxic fluorescent nucleic acid stain supplied as a DMSO solution. The dye is a unique oily macromolecule that cannot penetrate cell membranes and is not volatile, so it cannot be inhaled during routine handling. Ames test results confirm that it is completely non-mutagenic at gel staining concentrations. Because the stain shares the same spectral properties as ethidium bromide (EB), no change of filters or viewing equipment is required—simply detect with UV excitation at 300 nm on a standard UV gel transilluminator. It is suitable for staining dsDNA, ssDNA, and RNA in both agarose and polyacrylamide gels, using either the pre-cast (in-gel) or post-staining (soaking) method for maximum convenience and flexibility.
Intended Use Fluorescent staining of dsDNA, ssDNA, and RNA in agarose and polyacrylamide gel electrophoresis, as a safe alternative to ethidium bromide.
Principle / Technology A safe nucleic acid dye based on a unique oily macromolecule that cannot cross cell membranes and does not volatilize. It has the same spectral characteristics as EB and fluoresces upon binding to nucleic acids, serving as a direct EB replacement.
Detection Method Same spectral properties as EB; no change of filters or observation devices is required. Optimal excitation with UV light around 300 nm on a standard UV gel transilluminator. Note: UV excitation is required; blue-light imagers are not suitable.
Sample Type dsDNA, ssDNA, and RNA.
Performance Range / Specifications Sensitivity comparable to EB, enabling efficient electrophoretic staining. In a comparison with EB, 100 bp DNA ladder and 2000 DNA Marker were loaded at 2, 3, 4, and 5 µL per lane and run at 140 V for 40 min, showing equivalent staining performance.
Sensitivity / LOD Sensitivity comparable to ethidium bromide (EB).
Specificity Stains dsDNA, ssDNA, and RNA in agarose and polyacrylamide gels.
Reaction Conditions / Protocol Both pre-cast gel staining and post-staining (soaking) methods are supported. No pre-dilution of the stock is required for pre-cast staining: add the 10,000× stain directly to the gel solution at the recommended ratio so that it is diluted to 1× in the gel. Adding the dye directly into samples before loading is not recommended.
Components / Formulation 10,000× nucleic acid gel stain solution in DMSO.
Storage Conditions Store at 4°C, protected from light and kept dry.
Shelf Life 2 years when stored as recommended.
Package Specifications 500 μL
Product Form Liquid; 10,000× solution in DMSO.
Quality Control Each lot is verified for staining sensitivity equivalent to EB; Ames test results demonstrate no mutagenicity at gel staining concentrations.
Concentration 10,000×
Shipping Conditions Shipped with ice packs.
Expiration Date / Stability Stable for 2 years when stored at 4°C, protected from light and kept dry. Waste dye decomposes naturally in the environment and requires no special disposal treatment.
Regulatory / Compliance Ames test results show the dye is completely non-mutagenic at gel staining concentrations; it is non-carcinogenic at working concentration, cannot penetrate cell membranes, and is a safe, non-toxic alternative to ethidium bromide. Standard laboratory safety practices should still be followed.
Compatibility Identical spectral properties to EB—no change of filters or observation devices required; suitable for staining before or after agarose or polyacrylamide gel electrophoresis.
Application Notes / Precautions Non-toxic: the oily macromolecular structure cannot penetrate cell membranes and is not volatile, and Ames testing confirms no mutagenicity at staining concentrations; standard lab safety rules still apply.
Both pre-cast gel staining and post-staining are supported; do not add the dye directly to samples before loading.
Stains dsDNA, ssDNA, and RNA.
No special observation device is needed: same spectrum as EB, best excited near 300 nm UV; not compatible with blue-light imagers.
No pre-dilution required: add the stock directly into the gel solution, where it is diluted to 1×.
