| Product Name | Recombinant TEV Protease with Removable Polyhistidine Tag (5 U/uL, Nickel Affinity Purified) |
| Catalog No. | IVDRM-0298 |
| Description | Engineered TEV protease keeps the usefulness of the viral enzyme with better stability and specificity. Its recognition motif is long enough to be effectively unique, so digestion gives a single cut between the glutamine and glycine or serine of the Glu-Asn-Leu-Tyr-Phe-Gln-Gly sequence most laboratories insert. Peak activity is at pH 7.0 and 30 °C, but the enzyme stays functional from pH 6.0 to 8.5 and from 4 to 30 °C, so a temperature-sensitive target can be digested overnight in the cold at the cost of longer incubation. This preparation is purified by nickel affinity chromatography, which also documents the N-terminal polyhistidine tag it carries and therefore its removability after digestion. Activity data are published for 4, 16, 21 and 30 °C, so the digestion schedule can be planned from measured rather than assumed performance. |
| Intended Use | Removal of affinity tags from recombinant proteins, particularly where this read-out is used at lower temperatures to protect unstable targets. |
| Principle / Technology | Recognition of the seven-residue sequence followed by hydrolysis of the bond between glutamine and the following small residue. |
| Sample Type | Fusion proteins in solution carrying the recognition motif. |
| Performance Range / Specifications | 5 U/uL; at least 95% pure by SDS-PAGE; nickel affinity purified; published cleavage data across 4, 16, 21 and 30 °C; active from pH 6.0 to 8.5; removable on nickel resin. |
| Specificity | Specific activity 5 U/uL; purity at least 95% by SDS-PAGE; purification by nickel affinity chromatography; recognition sequence Glu-Asn-Leu-Tyr-Phe-Gln cleaved before glycine; offered as 1000 U, 10 x 1000 U, 10 KU and 50 KU. |
| Reaction Conditions / Protocol | Dilute the substrate into 1x reaction buffer of 50 mM Tris at pH 8.0 with 0.5 mM EDTA and 1 mM dithiothreitol, add the enzyme, and incubate at the chosen temperature for the time indicated by the published activity table. |
| Components / Formulation | Protease solution and 10x reaction buffer. |
| Storage Conditions | Protease at -20 °C for 6 months or below -80 °C for 1 year; buffer likewise. |
| Shelf Life | 6 months at -20 °C or 1 year below -80 °C. |
| Package Specifications | Supplied as 1000 U, 10 x 1000 U, 10 KU or 50 KU with matching buffer volumes. |
| Product Form | Colourless, clear liquid. |
| Quality Control | Released at not less than 95% purity by SDS-PAGE with unit activity confirmed against the standard substrate. |
| Key Features | - At least 95% pure by SDS-PAGE with nickel affinity purification - Published cleavage percentages at 4, 16, 21 and 30 °C for planning - Works from pH 6.0 to 8.5 for sensitive targets - Single cut at a seven-residue motif rarely found by chance - Removed on nickel resin through the polyhistidine tag |
| Purity | At least 95% by SDS-PAGE. |
| Concentration | 5 U/uL. |
| Activity / Unit Definition | One unit cleaves more than 85% of 3 ug of substrate in 1 hour at 30 °C in 1x reaction buffer. |
| Molecular Weight | Not restated on the source page beyond the purity figure. |
| Source / Origin | Recombinant expression in Escherichia coli with nickel affinity purification. |
| pH Range / Optimal pH | Optimum pH 7.0; functional from 6.0 to 8.5. |
| Shipping Conditions | Shipped with ice packs. |
| Expiration Date / Stability | Holds performance when kept below -80 °C; avoid repeated warming. |
| Regulatory / Compliance | Supplied for laboratory research purposes. |
| Compatibility | Removable on nickel resin because of the polyhistidine tag; the published temperature profile allows planning around second temperature-sensitive targets. |
| Recommended Buffer System | 1x reaction buffer is 50 mM Tris at pH 8.0 with 0.5 mM EDTA and 1 mM dithiothreitol, supplied as a 10x concentrate. |
| Application Notes / Precautions | Use published cleavage percentages rather than assuming low temperature equivalence: at 4 °C one hour clears far less substrate than at 30 °C. |
| Batch-to-Batch Consistency | Each production lot is released against fixed acceptance criteria so performance stays consistent between orders. |
| Model Type | Site-specific recombinant protease |
| Starting Material / Tissue Site | Recombinant protease from Escherichia coli with an N-terminal polyhistidine tag |
| Delivery Standard | Protease vial plus 10x buffer vial |
| Turnaround Time | No lead time is published on the product page; availability is confirmed when the order is placed. |
| Readouts / Downstream Assays | Fraction of substrate cleaved judged from the SDS-PAGE shift and from the published activity table. |
For in vitro diagnostic use.
