| Product Name | Recombinant TEV Protease, High-Purity Grade with Removable Polyhistidine Tag (5 U/uL) |
| Catalog No. | IVDRM-0297 |
| Description | TEV protease is the workhorse for removing affinity tags because its seven-residue recognition sequence almost never appears by accident in a target protein. This version is the result of a reworked production process: purity and activity are both higher than the standard preparation and residual host DNA is lower, which matters when the digested protein has to meet a tight residual specification. It recognises the heptapeptide Glu-Asn-Leu-Tyr-Phe-Gln and cuts between the glutamine and the following glycine or serine. Activity peaks at pH 7.0 and 30 °C, yet the enzyme still works across pH 6.0 to 8.5 and between 4 and 30 °C, so conditions can be tuned to the stability of the target. An N-terminal polyhistidine tag lets the protease itself be removed on nickel resin afterwards. |
| Intended Use | Cleavage of affinity tags from recombinant fusion proteins where higher purity and lower residual host DNA are required. |
| Principle / Technology | The protease recognises a seven-residue motif and hydrolyses the bond between the glutamine and a following small residue, giving single-site cutting. |
| Detection Method | Does not apply; performance is judged by the downstream assay rather than by this product. |
| Sample Type | Partially purified fusion proteins carrying the recognition sequence. |
| Performance Range / Specifications | Higher purity and stronger activity than the standard preparation; lower residual host DNA; active from pH 6.0 to 8.5 and from 4 to 30 °C; removable through the polyhistidine tag. |
| Specificity | 5 U/uL; purity at least 95%; recognition sequence Glu-Xaa-Xaa-Tyr-Xaa-Gln followed by glycine or serine; optimum at pH 7.0 and 30 °C; supplied as 1 KU, 10 KU, 50 KU or 1 MU (1000 KU). |
| Reaction Conditions / Protocol | Set up the digestion in 1x protease buffer containing 50 mM Tris at pH 8.0, 0.1 mM EDTA and 1 mM dithiothreitol, incubate at 30 °C for about an hour, then pass the mixture over nickel resin to remove the tagged protease. |
| Components / Formulation | Enzyme solution plus 10x concentrated reaction buffer. |
| Storage Conditions | Enzyme part at -85 to -65 °C for 1 year or -25 to -15 °C for 6 months; buffer part at -25 to -15 °C for 1 year. |
| Shelf Life | 1 year deep frozen for the enzyme part, 6 months at -25 to -15 °C; buffer part stable 1 year. |
| Package Specifications | Enzyme at 5 U/uL with matching 10x buffer; quantities of 1 KU, 10 KU, 50 KU or 1 MU (1000 KU). |
| Product Form | Clear liquid enzyme with separate concentrated buffer. |
| Quality Control | Released at not less than 95% purity with residual host DNA verified below the specification for this grade. |
| Key Features | - Refined process gives higher purity and activity than the standard preparation - Lower residual host DNA for sensitive applications - Cuts between glutamine and glycine or serine of the heptapeptide motif - Active from pH 6.0 to 8.5 and 4 to 30 °C - Removed afterwards on nickel resin through the polyhistidine tag |
| Purity | At least 95%. |
| Concentration | 5 U/uL. |
| Activity / Unit Definition | One unit cleaves more than 85% of 3 ug of substrate in 1 hour at 30 °C in the stated buffer. |
| Molecular Weight | Not restated on the source page beyond the purity specification. |
| Source / Origin | Recombinant expression in Escherichia coli. |
| pH Range / Optimal pH | Optimum pH 7.0 with measurable activity from 6.0 to 8.5. |
| Shipping Conditions | Shipped frozen. |
| Expiration Date / Stability | Stability depends on continuous storage at the stated temperature with the container closed. |
| Regulatory / Compliance | Restricted to research applications; not for clinical use. |
| Compatibility | Works with nickel affinity purification through the polyhistidine tag and with the companion 10x reaction buffer. |
| Recommended Buffer System | 1x reaction buffer is 50 mM Tris at pH 8.0 with 0.1 mM EDTA and 1 mM dithiothreitol, supplied as a 10x concentrate. |
| Application Notes / Precautions | Thaw aliquots rather than the whole stock, keep dithiothreitol in the reaction buffer unless the target is sensitive to reducing conditions, and remember that activity falls as temperature is reduced. |
| Batch-to-Batch Consistency | Fixed production controls and release testing keep performance uniform from lot to lot. |
| Model Type | Site-specific recombinant protease |
| Starting Material / Tissue Site | Recombinant protease from Escherichia coli carrying an N-terminal polyhistidine tag |
| Delivery Standard | Enzyme vial plus 10x buffer vial |
| Readouts / Downstream Assays | Shift of the target band on SDS-PAGE after digestion and residual host DNA in the final material. |
For in vitro diagnostic use.
