| Product Name | Recombinant DNase I (RNase-Free, Yeast-Expressed, 5 U/μL) |
| Catalog No. | IVDRM-0030 |
| Description | Recombinant DNase I (Deoxyribonuclease I) is an endonuclease that digests single-stranded and double-stranded DNA. Expressed in yeast and verified free of RNase contamination, it retains strong digestion activity even in complex sample environments, making it a reliable choice for removing DNA from RNA samples and other demanding nucleic acid workflows. In side-by-side testing against plasmid DNA, its DNA removal efficiency was comparable to leading competing products. |
| Intended Use | Removal of DNA during the processing of various RNA samples, including elimination of genomic DNA contamination in RNA extraction and related nucleic acid workflows. |
| Principle / Technology | DNase I hydrolyzes phosphodiester bonds in DNA, yielding mono- and oligodeoxynucleotides with 5'-phosphate and 3'-OH termini. Its activity is Ca2+-dependent and can be stimulated by divalent metal ions such as Co2+, Mn2+, and Zn2+. In the presence of Mg2+, DNase I cuts double-stranded DNA at random sites; in the presence of Mn2+, it cleaves both strands at the same position, generating blunt ends or sticky ends with 1–2 nucleotide overhangs. |
| Sample Type | DNA removal in the processing of various RNA samples; validated on plasmid DNA digestion and on mouse liver pre-treatment samples for RNA extraction. |
| Performance Range / Specifications | Recombinant enzyme expressed in yeast. No detectable RNase contamination. Strong digestion capacity: efficiently digests DNA even in complex sample environments. DNA removal efficiency comparable to leading competing products. Validated in RNA extraction: RNA integrity remained intact after treatment of 12 mouse liver pre-treatment samples from different sources with 20 U of the enzyme. |
| Specificity | Digests single-stranded and double-stranded DNA; no detectable RNase activity, ensuring RNA samples are not degraded. |
| Reaction Conditions / Protocol | Optimal working pH range is 7–8. Activity depends on Ca2+ and is stimulated by divalent metal ions (Co2+, Mn2+, Zn2+). With Mg2+, double-stranded DNA is cleaved at random sites; with Mn2+, both strands are cleaved at the same position to produce blunt ends or 1–2 nucleotide sticky ends. |
| Storage Conditions | Store at -25 to -15°C. |
| Shelf Life | 2 years when stored at -25 to -15°C. |
| Package Specifications | 1000 U 5000 U |
| Quality Control | RNase contamination test: 10 U of the enzyme was incubated with RNA substrate at 37°C for 1 h; agarose gel electrophoresis showed no detectable RNase activity in any of the three batches tested, eliminating the risk of RNA sample degradation. |
| Concentration | 5 U/μL |
| Source / Origin | Recombinant, expressed in yeast. |
| pH Range / Optimal pH | Optimal pH 7-8 |
| Expiration Date / Stability | Stable for 2 years at -25 to -15°C. |
| Batch-to-Batch Consistency | RNase contamination was verified across three independent production batches: after incubation with RNA substrate at 37°C for 1 h, no RNase activity was detected in any batch, demonstrating consistent RNase-free quality from batch to batch. |
