Description A synthetic polymer coating solution for tissue culture surfaces that promotes cell attachment through electrostatic interactions. Poly-D-lysine provides a positively charged surface suitable for a broad range of cell types.
Intended Use Enhancement of cell attachment to tissue culture plastic and glass surfaces for primary cells, neuronal cultures, transfected cells, and other cell types that require a positively charged substrate.
Principle / Technology Poly-D-lysine is a synthetic cationic polymer that adsorbs to negatively charged tissue culture surfaces, creating a positively charged monolayer; cells attach through electrostatic interactions between negatively charged cell surface proteoglycans and the poly-D-lysine coating.
Detection Method Cell attachment assessed by phase-contrast microscopy; cell spreading and proliferation monitored using standard culture observation techniques

Reaction Conditions / Protocol Dilute poly-D-lysine stock to 50–100 μg/mL in sterile water or PBS, add to culture vessel, incubate at room temperature for 1 hour or overnight at 2–8°C, aspirate excess solution, wash 2–3 times with sterile water, allow to air-dry in a sterile hood, seed cells
