Product Name PBCV DNA Ligase for RNA-Splinted Ligation and Target Enrichment (25 U/uL)
Catalog No. IVDRM-0431
Description Also known as SplintR ligase or Chlorella virus DNA ligase, this ATP dependent enzyme from phage PBCV-1 efficiently joins two adjacent DNA strands hybridised to a complementary RNA splint, with the acceptor strand providing the 3 prime hydroxyl and the donor strand the 5 prime phosphate. Its affinity for RNA splinted DNA substrates, apparent Km of 1 nM, far exceeds conventional T4 DNA ligase and enables sub nanomolar detection of distinct RNA species in complex mixtures. Established applications include padlock probe circularisation for in situ sequencing followed by phi29 driven rolling circle amplification, adjacent DNA ligation and target enrichment pipelines in next generation sequencing and molecular diagnostics.
Intended Use Padlock probe circularisation for in situ sequencing, RNA splinted ligation, RNA detection and target enrichment for NGS and molecular diagnostics
