| Product Name | Nt.BspQI Nicking Endonuclease (10 U/uL) |
| Catalog No. | IVDRM-0539 |
| Description | Unlike conventional endonucleases, Nt.BspQI cuts only one strand of a double stranded DNA substrate, introducing a nick at the 5 prime GCTCTTCN 3 prime site on the top strand while leaving the DNA molecule uncut through its full length. This supports molecular cloning workflows that need a defined site-specific single-strand nick. The enzyme runs in its dedicated buffer at 50°C and is heat inactivated at 80°C for 20 minutes, and one unit converts 1 microgram of supercoiled pUC19 DNA completely to the open circular form in one hour at 50°C in a 50 microlitre reaction. |
| Intended Use | Site-specific single-strand nicking of double stranded DNA for molecular cloning |
| Principle / Technology | Top strand cleavage at the recognition site that leaves the duplex intact |
| Sample Type | Supercoiled plasmid DNA containing the recognition site |
| Specificity | Nicks the top strand only without cutting the DNA molecule |
| Reaction Conditions / Protocol | Digest in 1x dedicated buffer at 50°C; heat inactivate at 80°C for 20 minutes |
| Storage Conditions | -25 to -15°C |
| Shelf Life | 2 years |
| Package Specifications | 2000 U |
| Key Features | Site-specific nicking at elevated 50°C digestion temperature |
