| Product Name | Nt.BbvCI Nicking Endonuclease (5 U/uL) |
| Catalog No. | IVDRM-0538 |
| Description | A nicking endonuclease that cuts only one strand of a double stranded DNA substrate, Nt.BbvCI introduces a nick on the top strand of the 5 prime CC^TCAGC 3 prime site without severing the duplex. Nicks produced by the enzyme trigger polymerase strand displacement in isothermal amplification methods such as SDA and RCA, and repeated nicking, displacement and extension drives exponential amplification of the target. The enzyme runs in the shared rapid series buffer at 37°C and is heat inactivated at 80°C for 20 minutes, and one unit converts 1 microgram of supercoiled p615 DNA completely to the open circular form in one hour at 37°C in a 50 microlitre reaction. |
| Intended Use | Single-strand nicking for molecular cloning and isothermal amplification methods such as SDA and RCA |
| Principle / Technology | Single strand cleavage on the top strand of the recognition site that seeds polymerase mediated strand displacement |
| Sample Type | Supercoiled plasmid DNA containing the recognition site |
| Specificity | Nicks the top strand only, leaving the duplex intact |
| Reaction Conditions / Protocol | Digest in 1x rapid restriction buffer at 37°C; heat inactivate at 80°C for 20 minutes |
| Storage Conditions | -25 to -15°C |
| Shelf Life | 2 years |
| Package Specifications | 150 U |
| Key Features | Nicks without cutting the duplex; triggers exponential isothermal amplification cycles |
| Concentration | 5 U/uL |
| Readouts / Downstream Assays | Conversion of supercoiled substrate to open circular form |
