| Product Name | NLS-Cas9-BPNLS Nuclease, Tag-Free, Dual Nuclear Localisation Signals |
| Catalog No. | IVDRM-0508 |
| Description | A tag-free Cas9 nuclease carrying the Streptococcus pyogenes Cas9 gene with a bipartite nuclear localisation signal at the N terminus and a nucleoplasmin nuclear localisation signal at the C terminus. The two signals raise nuclear delivery markedly and thereby improve editing efficiency. The protein forms a ribonucleoprotein complex with guide RNA, and RNP-based editing avoids problems seen with viral and plasmid delivery such as off-target effects and burdens on cell viability and transcription or translation. Editing is consistently high in vitro and in vivo, and the preparation contains no exogenous DNA. |
| Intended Use | In vitro and in vivo genome editing through Cas9 RNP complexes with reduced off-target effects |
| Principle / Technology | Guide RNA directed Cas9 nuclease delivered as an RNP complex, with dual nuclear localisation signals supporting nuclear entry |
| Sample Type | Cultured cells transfected with the Cas9 RNP complex |
| Specificity | N-terminal bipartite BPNLS and C-terminal nucleoplasmin NLS; consistently high editing efficiency in vitro and in vivo |
| Reaction Conditions / Protocol | Combine with guide RNA to form the RNP complex, then deliver into cells by transfection or electroporation |
| Storage Conditions | -25 to -15°C |
| Shelf Life | 1 year |
