| Product Name | Nb.BbvCI Nicking Endonuclease (10 U/uL) |
| Catalog No. | IVDRM-0536 |
| Description | A nicking endonuclease that cuts only one strand of a double stranded DNA substrate, Nb.BbvCI introduces a nick at its recognition site without severing the duplex. Nicks produced by the enzyme trigger polymerase strand displacement reactions in isothermal amplification methods such as SDA and RCA, and repeated nicking, displacement and extension drives exponential amplification of the nucleic acid target. The enzyme runs in the shared rapid series buffer at 37°C and is heat inactivated at 80°C for 20 minutes. EcoBI methylation overlapping the site blocks cleavage. |
| Intended Use | Generation of single-strand nicks for strand displacement amplification and rolling circle amplification |
| Principle / Technology | Single strand cleavage at the recognition site that seeds polymerase mediated strand displacement |
| Sample Type | Supercoiled plasmid DNA or amplification substrates containing the recognition site |
| Specificity | Nicks one strand only; EcoBI methylation overlapping the site blocks cleavage |
| Reaction Conditions / Protocol | Digest in 1x rapid restriction buffer at 37°C; heat inactivate at 80°C for 20 minutes |
| Storage Conditions | -25 to -15°C |
| Shelf Life | 2 years |
| Package Specifications | 300 U |
| Key Features | Nicks without cutting the duplex; triggers exponential isothermal amplification cycles |
