Description A 100-fold concentrated chemically defined serum-free supplement based on Bottenstein's N2 formula, containing insulin, transferrin, progesterone, putrescine, and selenite for neural stem cell and neural crest cell culture.
Intended Use Supplementing DMEM/F12 medium for the serum-free culture of neural stem cells, neuroblastoma lines, and neural crest-derived cells; component of defined neuronal differentiation media for pluripotent stem cells.
Principle / Technology Insulin promotes glucose uptake and cell survival signaling through IGF-1 receptor; transferrin provides iron for enzymatic function; progesterone supports myelination; putrescine promotes cell proliferation; selenite is a cofactor for glutathione peroxidase antioxidant defense.
Detection Method Nestin and Sox2 expression by immunocytochemistry, neurosphere formation assay, differentiation capacity into neurons, astrocytes, and oligodendrocytes
