| Product Name | Mutant Taq DNA Ligase for Homologous Recombination and Seamless Cloning (40 U/uL) |
| Catalog No. | IVDRM-0501 |
| Description | A mutant form of Taq DNA Ligase with markedly higher ligation efficiency that improves performance in homologous recombination workflows while retaining the parent enzyme functions. Like the wild type, this thermostable ligase joins the 5 prime phosphate and 3 prime hydroxyl ends of two oligonucleotides hybridised in adjacent positions on the same complementary target strand, and the reaction proceeds only when both oligonucleotides are perfectly paired and gap free, so single base substitutions can be detected in ligase detection and ligase chain reaction formats. The NAD+ dependent enzyme remains active between 45 and 65°C and also serves mutagenesis by incorporation of phosphorylated oligonucleotides during PCR amplification. |
| Intended Use | Homologous recombination and seamless cloning, allele-specific detection and PCR-based mutagenesis |
| Principle / Technology | NAD-dependent, gap-free ligation of adjacent perfectly paired oligonucleotides at 45 to 65°C with enhanced efficiency |
| Sample Type | DNA fragments with homologous ends; oligonucleotide probes on a complementary target |
| Specificity | Mutant enzyme with higher ligation efficiency; ligates only perfectly paired adjacent oligonucleotides with no gap |
| Reaction Conditions / Protocol | Incubate with the assembly or detection substrate at 45 to 65°C in an NAD+ containing buffer |
| Storage Conditions | -20°C; store the buffer at -80°C for periods beyond 30 days |
| Shelf Life | 2 years |
