| Product Name | Low Melting Point Agarose |
| Catalog No. | IVDRM-0114 |
| Description | Low melting point agarose is a modified agarose with a lower gelling temperature and melting point than standard agarose. It offers improved molecular sieving characteristics and sharper band resolution. It is ideally suited for separating nucleic acids larger than 1000 bp and for recovering nucleic acid fragments after electrophoresis, since it melts at approximately 65.5°C—below the melting point of virtually all nucleic acid molecules. DNA bands purified and recovered from low melting point agarose gels can be remelted and used directly to prepare radioisotope-labeled DNA probes. Restriction fragments separated on low melting point agarose gels can also be conveniently cloned directly into plasmids after gel excision and remelting. The product is additionally suitable for cloning of tissue culture cells and for viral plaque assays. |
| Intended Use | Separation of nucleic acids larger than 1000 bp; recovery of nucleic acid fragments after electrophoresis; direct preparation of radioisotope-labeled DNA probes from remelted gel slices; direct cloning of restriction fragments into plasmids; cloning of tissue culture cells; viral plaque assays; soft agar assays such as tumor soft agar colony formation assays. |
| Principle / Technology | Chemical modification lowers the gelling temperature and melting point of the agarose (melting point approximately 65.5°C), allowing gel slices containing recovered nucleic acids to be remelted at temperatures that do not denature DNA. The modified matrix also provides better molecular sieving performance and clearer bands. Standard agarose quality parameters—sulfate content, gel strength, gelling point, and electroendosmosis (EEO)—remain applicable. |
| Sample Type | DNA and RNA samples; also used with tissue culture cells in cloning and viral plaque assays. |
| Performance Range / Specifications | Melting point: approximately 65.5°C, below the melting point of virtually all nucleic acid molecules Recommended separation range: nucleic acids > 1000 bp Superior molecular sieving characteristics and higher band clarity Key matrix parameters: sulfate content, gel strength, gelling point, electroendosmosis (EEO) |
| Reaction Conditions / Protocol | Prepare gels at the desired concentration in electrophoresis buffer. For fragment recovery, excise the band of interest, remelt the gel slice at approximately 65.5°C, and use the molten solution directly in downstream enzymatic reactions such as probe labeling or ligation. |
| Components / Formulation | Modified low melting point agarose. |
| Storage Conditions | Store at room temperature. |
| Shelf Life | 5 years. |
| Package Specifications | 5 g; 25 g; 100 g |
| Product Form | Powder |
| Quality Control | Sterile and free of DNase/RNase; suitable for cell-based applications such as viral plaque assays and tissue culture. |
| Purity | Sulfate content is monitored as the purity indicator, as sulfate is the major ionic group present in agarose. |
| Expiration Date / Stability | Stable for 5 years when stored at room temperature. |
| Compatibility | Recovered DNA can be used directly for radioisotope-labeled probe preparation and direct ligation/cloning into plasmids after gel remelting. Compatible with soft agar colony formation assays and viral plaque assays. Not suitable for pulsed-field gel electrophoresis, which requires very low EEO and high gel strength. |
| Application Notes / Precautions | Suitable for soft agar applications, including tumor soft agar colony formation assays. Gel slices can be remelted at approximately 65.5°C and used directly in downstream enzymatic reactions without further purification. |
For research use only, not for clinical use.
