Description A defined serum-free supplement designed to replace fetal bovine serum in the culture of mouse and human embryonic stem cells and induced pluripotent stem cells while maintaining pluripotency and normal karyotype.
Intended Use Replacing FBS in ES and iPS cell culture medium to maintain undifferentiated growth, genomic stability, and pluripotency without the differentiation-inducing effects and lot-to-lot variability of serum.
Principle / Technology Contains amino acids, vitamins, transferrin, insulin, selenium, and lipid-rich BSA in a formulation that supports self-renewal signaling through LIF/STAT3 (mouse) and FGF2/TGF-beta (human) pathways without serum-induced differentiation signals.
Detection Method Alkaline phosphatase staining for pluripotency, Oct4/Nanog/Sox2 expression by immunostaining, karyotype analysis

Reaction Conditions / Protocol Supplement KnockOut DMEM or DMEM/F12 with 15–20% KnockOut Serum Replacement, L-glutamine, NEAA, and 2-mercaptoethanol; add 1000 U/mL LIF for mouse ES cells or 4–10 ng/mL bFGF for human ES cells.
