| Product Name | I-SceI Homing Endonuclease (5 U/uL) |
| Catalog No. | IVDRM-0541 |
| Description | An intron-encoded endonuclease from Saccharomyces cerevisiae, I-SceI specifically recognises and cuts the 18 base pair non-palindromic sequence 5 prime TAGGGATAA^CAGGGTAAT 3 prime, leaving a four base 3 prime sticky overhang. Because the recognition sequence is extremely long, it occurs naturally in genomes only about once in every seven times ten to the tenth random bases, which makes the enzyme an ideal tool for precise genome operations and targeted genomic editing. It runs in the shared rapid series buffer at 37°C and is heat inactivated at 80°C for 20 minutes, with one unit digesting 1 microgram of pUC-I-SceI substrate in one hour at 37°C Methylation has no effect on cleavage. |
| Intended Use | Precise genome editing and genomic operations through rare 18 bp site cleavage |
| Principle / Technology | Recognition and cleavage of an extremely long non-palindromic 18 base pair site producing a four base 3 prime overhang |
| Sample Type | Genomic DNA or engineered constructs carrying the I-SceI site |
| Specificity | Cuts a 18 bp non-palindromic site with a four base 3 prime overhang; unaffected by methylation |
| Reaction Conditions / Protocol | Digest in 1x rapid restriction buffer at 37°C; heat inactivate at 80°C for 20 minutes |
| Storage Conditions | -25 to -15°C |
| Shelf Life | 2 years |
| Package Specifications | 250 U |
| Key Features | Extremely rare natural occurrence of the site; suited to precise genome targeting |
| Concentration | 5 U/uL |
| Readouts / Downstream Assays | Agarose gel profile of digested DNA |
For in vitro diagnostic use.
