| Product Name | High-Sieving Agarose (PCR Grade) |
| Catalog No. | IVDRM-0115 |
| Description | Agarose is a purified linear galactan hydrocolloid obtained from agar or agar-bearing seaweed, structured as a linear polymer of β-D-galactopyranose (1→4)-linked to 3,6-anhydro-α-L-galactopyranose. This high-sieving, PCR-grade agarose is optimized for fine resolution of PCR products and small DNA fragments, with a recommended separation range of 20 bp to 800 bp and a size resolution of approximately 2%. It dissolves easily to give clear gel solutions, can be cast at concentrations up to 5%, produces very low background, and delivers consistent quality from lot to lot. |
| Intended Use | High-resolution separation of PCR products and small DNA fragments in the 20 bp–800 bp range; routine nucleic acid analysis by gel electrophoresis and blotting techniques such as Northern and Southern blotting. |
| Principle / Technology | The high-sieving agarose matrix provides enhanced molecular sieving for small DNA fragments, achieving approximately 2% size resolution (e.g., a 100 bp fragment can be resolved from fragments differing by about 2 bp). Gels can be prepared at concentrations up to 5% to further improve resolution of small fragments. Key matrix parameters include sulfate content, gel strength, gelling point, and electroendosmosis (EEO). |
| Sample Type | PCR products and small DNA fragments (20 bp–800 bp). |
| Performance Range / Specifications | Separation range: 20 bp–800 bp DNA fragments Size resolution: approximately 2% Maximum gel concentration: up to 5% Gel strength: ≥ 1200 g/cm² Easy to dissolve; clear gel solutions; very low background Stable, consistent quality |
| Reaction Conditions / Protocol | Dissolve in electrophoresis buffer by heating and cast at the desired concentration, up to 5% for maximal small-fragment resolution. For a 1% gel, allow a minimum gelling time of 30 minutes to ensure a homogeneous gel and sharp bands. |
| Components / Formulation | Purified high-sieving linear galactan polysaccharide extracted from agar or agar-bearing seaweed. |
| Storage Conditions | Store dry at room temperature. |
| Shelf Life | 5 years. |
| Package Specifications | 5 g; 25 g; 100 g |
| Product Form | Powder |
| Quality Control | Gel strength is verified to be ≥ 1200 g/cm². Electrophoretic performance is validated by resolution of small DNA fragments at approximately 2% size resolution. A 1% gel requires a minimum gelling time of 30 minutes for uniform, well-resolved bands. |
| Purity | Sulfate content is monitored as the purity indicator, as sulfate is the major ionic group present in agarose; gels exhibit very low background. |
| Expiration Date / Stability | Stable for 5 years when stored dry at room temperature. |
| Compatibility | Suitable for analysis of PCR products and small DNA fragments; compatible with standard gel electrophoresis and blotting workflows such as Northern and Southern blotting. |
| Application Notes / Precautions | Diffuse bands are usually caused by insufficient gelling time or a non-uniform gel; allow at least 30 minutes for a 1% gel to solidify. Size resolution is approximately 2% (e.g., a 30 bp fragment can be resolved from fragments differing by about 0.6 bp; a 100 bp fragment from those differing by about 2 bp). Gel firmness is related to gel strength; as a natural agar extract, strength may vary slightly between batches, and all lots with strength ≥ 1200 g/cm² meet specification. |
| Batch-to-Batch Consistency | Consistent quality from lot to lot; gel strength may vary slightly as agarose is a natural agar extract, and all released lots meet the ≥ 1200 g/cm² specification. |
